嗎啡調(diào)節(jié)大鼠催產(chǎn)素合成與釋放的機制及GIRK在嗎啡效應中作用的研究

長期應用阿片可引起機體對阿片的依賴和耐受,但其機制目前尚不清楚, 這是神經(jīng)科學工作者面臨的重要研究課題之一。研究表明,下丘腦合成的神經(jīng) 垂體激素催產(chǎn)素(OT)參與藥物成癮和學習記憶過程,減弱外周和中樞注射嗎 啡引起的依賴和耐受,并減輕大鼠對海洛因的耐受和海洛因的自身給藥行為. 側(cè)腦室或邊緣前腦內(nèi)注射OT受體拮抗劑可易化嗎啡的耐受,提示中樞OT可 拮抗嗎啡的依賴和耐受的形成。急性嗎啡處理可抑制大鼠下丘腦視上核(SON) 神經(jīng)元的放電,并明顯抑制OT的釋放;慢性嗎啡處理,可導致中樞OT含量 降低,戒斷期間血漿OT水平出現(xiàn)瞬間的大量增加,表明中樞OT含量的變化 與嗎啡依賴的形成有密切關系。阿片抑制視上核OT的釋放,減輕OT對阿片 依賴和耐受的拮抗作用,從而易化阿片依賴和耐受的形成。因此闡明阿片抑制 視上核OT陽性神經(jīng)元及OT釋放的機制,對設計抗嗎啡依賴和耐受形成的藥 物有重要意義。...
