應激對SAMP8小鼠認知功能及情緒的影響及兩者機制研究

【摘要】:目的研究慢性不可預知溫和應激(UCMS)對SAMP8小鼠抑郁情緒、學習記憶能力的影響,及UCMS對海馬中腦源性神經(jīng)營養(yǎng)因子(BDNF)及其受體(Trk B)、神經(jīng)元凋亡相關(guān)因子B淋巴細胞瘤-2基因(Bcl-2)、Bcl-2相關(guān)X蛋白(Bax)蛋白表達的影響。方法選取健康、7月齡、雄性SAMP8小鼠20只,對其進行隨機分組,分為應激組和非應激組,每組各10只。然后對兩組小鼠進行為期21天的慢性不可預知應激,應激完成后再進行水迷宮實驗及糖水偏好試驗。行為學實驗完成后,應激組和非應激組各隨機選取6只小鼠進行斷頭取海馬,采用蛋白質(zhì)印跡法(westernblotting)對BDNF及Trk B、Bcl-2、Bax的蛋白表達量進行測定。結(jié)果1經(jīng)過5天的訓練后,應激組小鼠尋找到平臺的潛伏期明顯增長(P0.05);在第6天撤除平臺后,應激組小鼠穿越平臺次數(shù)比非應激組明顯減少(P0.05)。2應激后應激組小鼠比非應激組糖水消耗量明顯下降(P0.05)。3與非應激組比較,應激組小鼠BDNF及Trk B蛋白表達明顯減少,且具有統(tǒng)計學意義(P0.05)。4與非應激組比較,應激組小鼠海馬Bcl-2蛋白表達明顯減少,Bax蛋白表達明顯增多,且具有統(tǒng)計學意義(P0.05)。結(jié)論應激使SAMP8小鼠產(chǎn)生抑郁情緒、學習記憶能力下降;應激導致的BDNF及其Trk B、Bcl-2蛋白表達量減少,Bax蛋白量表達增多。
[Abstract]:Objective to study the effects of chronic unpredictable mild stress (UCMS) on depression, learning and memory ability in SAMP8 mice, and the effect of UCMS on brain-derived neurotrophic factor (BDNF) and its receptor (Trk B), in hippocampus. Effect of apoptosis-related factor B lymphocyte tumor-2 gene (Bcl-2) and Bcl-2-associated X protein (Bax) protein expression. Methods Twenty healthy 7-month-old male SAMP8 mice were randomly divided into two groups: stress group (n = 10) and non-stress group (n = 10). Two groups of mice were subjected to chronic unpredictable stress for 21 days. After the stress was completed, the water maze test and sugar-water preference test were performed. After the behavioral experiment was completed, six mice in the stress group and the non-stress group were randomly selected and the hippocampus was decapitated. The protein expression of BDNF, Trk-B, Bcl-2 and Bax were measured by protein blot (westernblotting). Results (1) after 5 days of training, the latency of finding the platform in the stress group increased significantly (P0.05). After removing the platform on the 6th day, the number of crossing the platform in the stress group was significantly lower than that in the non-stress group (P0.05). 2 the consumption of sugar water in the stress group was significantly lower than that in the non-stress group (P0.05). 3Compared with the non-stress group, the frequency of crossing the platform in the stress group decreased significantly (P0.05). (4) compared with non-stress group, the expression of BDNF and Trk B protein in hippocampus of stress group decreased significantly, and the expression of Bax protein increased significantly, compared with non-stress group, the expression of Bcl-2 and Bax protein in hippocampus of stress group was significantly decreased, and the expression of Bax protein was significantly higher than that of non-stress group. And has statistical significance (P0.05). Conclusion stress induced depression and decreased the ability of learning and memory in SAMP8 mice, the expression of Bcl-2 protein and Bcl-2 protein decreased and the expression of Bax protein increased in BDNF and Trk-B induced by stress.
