鷹嘴豆芽素A對過氧化氫誘導大鼠原代海馬神經(jīng)元損傷的作用及抗凋亡機制

【摘要】:目的探討植物雌激素鷹嘴豆芽素A對H_2O_2誘導大鼠原代海馬神經(jīng)元細胞凋亡的保護作用。方法取新生24 h內的海馬神經(jīng)元細胞,培養(yǎng)10 d左右,鑒定神經(jīng)細胞元純度。用雌激素和低、中、高劑量的鷹嘴豆芽素A處理細胞24 h,除正常組外,其余組給予H_2O_2作用12 h,CCK-8法測定細胞活力,用流式細胞儀測定各組細胞凋亡率。Western blot測定Bcl-2、Bax、caspase-3蛋白表達水平。結果用神經(jīng)元特異性抗體微管相關蛋白2熒光檢測海馬神經(jīng)元純度達98%以上,鷹嘴豆芽素A預處理24 h能顯著改善H_2O_2作用12 h后的海馬神經(jīng)元活力。用流式細胞儀觀察H_2O_2損傷后,凋亡細胞數(shù)和壞死細胞數(shù)明顯增加,給予鷹嘴豆芽素A預處理后,可明顯減少H_2O_2引起的凋亡和壞死細胞數(shù)。鷹嘴豆芽素A各劑量組劑量依賴性地提高模型細胞Bcl-2蛋白表達,降低Bax、caspase-3蛋白表達。結論鷹嘴豆芽素A對H_2O_2誘導大鼠原代海馬神經(jīng)元損傷的保護作用可能與其抑制細胞凋亡有關,鷹嘴豆芽素A抗神經(jīng)元凋亡作用的機制可能與其上調Bcl-2蛋白表達和下調Bax、caspase-3蛋白表達有關。
[Abstract]:Objective to investigate the protective effect of phytoestrogenin A on apoptosis of primary hippocampal neurons induced by H _ 2O _ 2 in rats. In addition to the normal group, the other groups were treated with H _ 2O _ 2 for 12 h to determine the cell viability by CCK-8 method. Apoptosis rate was measured by flow cytometry. Western blot was used to detect the expression of Bcl-2Baxax-caspase-3 protein. Results the purity of hippocampal neurons was more than 98% by fluorescence detection of neuron specific antibody microtubule-associated protein 2. The hippocampal neuronal activity of H2O2 was significantly improved after H2O2 was pretreated for 24 h. The number of apoptotic cells and necrotic cells increased significantly after H _ 2O _ 2 injury was observed by flow cytometry, and the number of apoptotic cells and necrotic cells were observed by flow cytometry. The number of apoptotic and necrotic cells induced by H _ 2O _ 2 was significantly reduced. The expression of Bcl-2 protein in model cells was increased in a dose-dependent manner. Conclusion the protective effect of chickpein A on the injury of primary hippocampal neurons induced by S _ 2O _ 2 in rats may be related to its inhibition of apoptosis. The mechanism of antiapoptotic effect of chickpein A may be related to its up-regulation of Bcl-2 protein expression and down-regulation of Bax-caspase-3 protein expression.
