銀杏內(nèi)酯B和鹿茸多肽對(duì)膽堿能前體細(xì)胞發(fā)生分化的研究

【摘要】:目的研究銀杏內(nèi)酯B和鹿茸多肽對(duì)膽堿能前體細(xì)胞發(fā)生、分化的影響,為膽堿能神經(jīng)元退行性疾病的治療探索和尋求新策略。方法實(shí)驗(yàn)在吉林大學(xué)白求恩醫(yī)學(xué)院組織胚胎學(xué)系細(xì)胞培養(yǎng)室和免疫組化實(shí)驗(yàn)室完成。原代培養(yǎng)E14-15大鼠端腦細(xì)胞,分別在實(shí)驗(yàn)組培養(yǎng)液中加入銀杏內(nèi)酯B、鹿茸多肽及兩者聯(lián)合,培養(yǎng)細(xì)胞4d后進(jìn)行乙酰膽堿轉(zhuǎn)移酶(ChAT)、5-溴尿嘧啶核苷(BrdU)免疫細(xì)胞化學(xué)染色,分別計(jì)算各組ChAT和BrdU免疫反應(yīng)陽(yáng)性細(xì)胞數(shù)。結(jié)果銀杏內(nèi)酯B、鹿茸多肽及聯(lián)合用藥組分別與對(duì)照組相比,神經(jīng)球較大,突起多而粗,且神經(jīng)球周邊細(xì)胞遷移數(shù)量多。ChAT免疫陽(yáng)性反應(yīng)細(xì)胞多分布于神經(jīng)球邊緣,神經(jīng)球之間可見(jiàn)散在陽(yáng)性細(xì)胞。藥物實(shí)驗(yàn)組ChAT免疫反應(yīng)陽(yáng)性細(xì)胞數(shù)量較對(duì)照組有顯著性增加,以聯(lián)合用藥組增加最為顯著,其次為鹿茸多肽組;Brdu免疫反應(yīng)陽(yáng)性細(xì)胞與對(duì)照組相比,鹿茸多肽組增加最為顯著。結(jié)論:銀杏內(nèi)酯B、鹿茸多肽可誘導(dǎo)端腦膽堿能前體細(xì)胞增殖和分化,聯(lián)合用藥作用效果更為顯著。
[Abstract]:Objective to study the effects of ginkgolide B and antler polypeptide on the development and differentiation of cholinergic precursor cells. Methods the experiment was carried out in the cell culture room and immunohistochemistry laboratory of the Department of Embryology of Bai Qiuen College of Medicine of Jilin University. The primary culture of E14-15 rat brain cells was carried out. Ginkgolide B, velvet antler polypeptide and their combination were added to the culture medium of the experimental group respectively. After cultured for 4 days, the cells were stained with acetylcholine transferase (ChATB) 5-brominouracil (BrdU) immunocytochemical staining. Results compared with the control group, Ginkgolide B, velvet antler polypeptide and the combination treatment group were larger than those in the control group, and the neurites were more and thicker than those in the control group, and the number of ChAT and BrdU immunoreactive positive cells in each group was calculated respectively. The number of ChAT immunoreactive cells in the experimental group was significantly higher than that in the control group. The increase of Brdu immunoreactive cells in the antler polypeptide group was higher than that in the control group. Conclusion: ginkgolide B and pilose antler polypeptide can induce the proliferation and differentiation of terminal cholinergic precursor cells.
