間充質(zhì)干細(xì)胞條件培養(yǎng)液對(duì)正常成纖維細(xì)胞及瘢痕成纖維細(xì)胞轉(zhuǎn)化生長(zhǎng)因子β產(chǎn)生和信號(hào)通路的影響

【摘要】:背景:已有文獻(xiàn)報(bào)道,間充質(zhì)干細(xì)胞可通過分泌大量生物活性因子有效減輕損傷的心臟、肺臟、腎臟等器官纖維化的發(fā)生。目的:觀察骨髓間充質(zhì)干細(xì)胞條件培養(yǎng)液對(duì)正常成纖維細(xì)胞和瘢痕成纖維細(xì)胞的影響及作用機(jī)制。方法:制備骨髓間充質(zhì)干細(xì)胞條件培養(yǎng)液,將其作用于正常成纖維細(xì)胞和增生性瘢痕成纖維細(xì)胞,采用ELISA檢測(cè)轉(zhuǎn)化生長(zhǎng)因子β的分泌水平,RT-PCR檢測(cè)TGF-β/Smad信號(hào)通路相關(guān)基因Smad7的表達(dá),比色法檢測(cè)各組細(xì)胞上清液中羥脯氨酸的含量。結(jié)果與結(jié)論:(1)骨髓間充質(zhì)干細(xì)胞條件培養(yǎng)液對(duì)增生性瘢痕成纖維細(xì)胞轉(zhuǎn)化生長(zhǎng)因子β的分泌、膠原的合成有顯著抑制作用(P0.01),并且明顯增強(qiáng)Smad7基因的表達(dá)(P0.01);(2)骨髓間充質(zhì)干細(xì)胞條件培養(yǎng)液對(duì)正常成纖維細(xì)胞的影響較小;(3)結(jié)果表明,骨髓間充質(zhì)干細(xì)胞條件培養(yǎng)液對(duì)增生性瘢痕成纖維細(xì)胞具有顯著抑制作用,為干細(xì)胞防治皮膚瘢痕提供基礎(chǔ)理論支持。
[Abstract]:Background: it has been reported that mesenchymal stem cells can effectively reduce the occurrence of fibrosis in injured heart, lung, kidney and other organs by secreting a large number of bioactive factors. Aim: to observe the effect and mechanism of conditioned medium of bone marrow mesenchymal stem cells on normal fibroblasts and scar fibroblasts. Methods: the conditioned medium of bone marrow mesenchymal stem cells was prepared and applied to normal fibroblasts and hypertrophic scar fibroblasts. The secretion of transforming growth factor 尾 (TGF 尾) was detected by ELISA. The expression of Smad7 related to TGF- 尾 / Smad signaling pathway was detected by RT-PCR, and the content of hydroxyproline in the supernatant of each group was detected by colorimetric assay. Results and conclusion: (1) conditioned medium of bone marrow mesenchymal stem cells significantly inhibited the secretion of TGF-尾 and collagen synthesis in hypertrophic scar fibroblasts (P0.01). And significantly increased the expression of Smad7 gene (P0.01); (2) the conditioned medium of bone marrow mesenchymal stem cells had little effect on normal fibroblasts; (3) the results showed that the conditioned medium of bone marrow mesenchymal stem cells had significant inhibitory effect on hypertrophic scar fibroblasts, which provided the basic theory support for the prevention and treatment of skin scar by stem cells.
