重度燒傷鼠腸上皮細(xì)胞及其刷狀緣谷氨酰胺轉(zhuǎn)運(yùn)的變化

【摘要】:目的探討燒傷后腸上皮細(xì)胞及其刷狀緣谷氨酰胺(glutamine,Gln)轉(zhuǎn)運(yùn)變化規(guī)律。方法將40只Sprague-Dawley大鼠隨機(jī)分為正常對(duì)照組(8只)、燒傷組(燒傷后1、3、5、7 d,每個(gè)時(shí)相點(diǎn)8只)。正常對(duì)照組只麻醉和剃毛,不予燒傷;燒傷組給予90℃水燙傷18 s,造成30%體表面積Ⅲ度燒傷。2組給予正常飲食及飲水。聯(lián)合應(yīng)用膠原酶Ⅸ和中性蛋白酶Ⅰ消化法提取大鼠小腸上皮細(xì)胞(intestinal epithelial cells,IEC),采用Mg2+差速離心法制備大鼠小腸刷狀緣膜囊泡(brush border membrane vesicles,BBMV),檢測(cè)進(jìn)入BBMV及IEC中的[3H]-Gln的放射性強(qiáng)度,觀察傷前及燒傷后1、3、5、7 d大鼠腸道BBMV及IEC中Gln轉(zhuǎn)運(yùn)速率的變化。結(jié)果采用Mg2+差速離心法成功提取大鼠腸上皮BBMV。發(fā)現(xiàn)腸上皮細(xì)胞中Na+依賴性Gln轉(zhuǎn)運(yùn)主要依靠BBMV,大約占總量的60%;燒傷第1天BBMV中Gln轉(zhuǎn)運(yùn)效率明顯下降(P0.05,P0.01),傷后3~7 d有所增高,但差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。IEC中Na+依賴性Gln轉(zhuǎn)運(yùn)變化不明顯,而非Na+依賴性Gln轉(zhuǎn)運(yùn)則有逐步增高的趨勢(shì),在傷后7 d明顯高于對(duì)照組(P0.05)。結(jié)論腸上皮細(xì)胞對(duì)Gln的轉(zhuǎn)運(yùn)主要依靠BBMV中Na+依賴性轉(zhuǎn)運(yùn),燒傷后BBMV轉(zhuǎn)運(yùn)Gln的能力先降低后增高,而IEC對(duì)非Na+依賴性Gln轉(zhuǎn)運(yùn)則呈逐步增高的趨勢(shì)。
