通過(guò)蛋白質(zhì)組學(xué)分析探究microRNA-21a-5p在小鼠脊髓損傷模型中的作用及相關(guān)信號(hào)通路

脊髓損傷(SCI)是脊柱損傷最嚴(yán)重的并發(fā)癥,是一種嚴(yán)重的神經(jīng)系統(tǒng)功能障礙性疾病,損傷后會(huì)導(dǎo)致?lián)p傷節(jié)段以下肢體發(fā)生嚴(yán)重的功能障礙,最典型的表現(xiàn)為損傷部位以下癱瘓,這不僅會(huì)給損傷患者本人帶來(lái)嚴(yán)重的身心傷害,還會(huì)給家庭和社會(huì)帶來(lái)沉重的經(jīng)濟(jì)、心理負(fù)擔(dān)。隨著現(xiàn)代社會(huì)經(jīng)濟(jì)的快速發(fā)展,尤其交通事故、工程建設(shè)和自然災(zāi)害等各類創(chuàng)傷的發(fā)病率上升,呈現(xiàn)致傷因子高能量、瞬間作用于人體造成多部位和多臟器損傷的特點(diǎn),脊髓損傷發(fā)生率出現(xiàn)逐年增高的趨勢(shì),盡管國(guó)內(nèi)外關(guān)于SCI的基礎(chǔ)、臨床研究眾多,但如何對(duì)脊髓損傷進(jìn)行有效的預(yù)防和治療已經(jīng)成為當(dāng)今醫(yī)學(xué)界的一大難題。microRNAs(miRs)是一類進(jìn)化上非常保守,長(zhǎng)度介于21~25nt的非編碼RNA(non-coding RNA,ncRNA)單鏈小分子,它能夠通過(guò)降解靶基因mRNA或者抑制其翻譯過(guò)程,達(dá)到負(fù)向調(diào)控目的蛋白的作用。miRs在不同生物體內(nèi)與靶標(biāo)結(jié)合的方式不盡相同,而且通?膳c多種蛋白相互作用。這種復(fù)雜的調(diào)節(jié)網(wǎng)絡(luò)既可以通過(guò)一個(gè)miR來(lái)調(diào)控多個(gè)基因的表達(dá),也可以聯(lián)合幾個(gè)miRs來(lái)精細(xì)調(diào)控某個(gè)基因的表達(dá)。miRs的這種特性決定了其可以同時(shí)調(diào)節(jié)多種靶蛋白,進(jìn)而調(diào)控細(xì)...
