弓形蟲(chóng)致密顆粒蛋白14基因的克隆及序列分析

【摘要】:為研究弓形蟲(chóng)致密顆粒蛋白14(GRA14)基因的遺傳變異規(guī)律,以來(lái)自不同宿主及地理來(lái)源的9株弓形蟲(chóng)DNA為模板,分別用PCR擴(kuò)增了其GRA14基因,并將PCR產(chǎn)物克隆到pMD18-T載體后測(cè)序。利用生物信息學(xué)軟件分析了GRA14基因的遺傳變異并構(gòu)建了系統(tǒng)發(fā)育樹(shù),分析了GRA14蛋白的結(jié)構(gòu)并結(jié)合抗原指數(shù)等參數(shù)預(yù)測(cè)了其抗原表位。結(jié)果表明,9個(gè)弓形蟲(chóng)蟲(chóng)株的GRA14基因長(zhǎng)度均為1 227bp,序列的A+T含量在46.94%~47.43%之間,與弓形蟲(chóng)ME49株相應(yīng)序列的核苷酸變異率在0.08%~0.73%之間;系統(tǒng)發(fā)育樹(shù)分析顯示,GRA14僅能將基因Ⅰ型蟲(chóng)株聚在一個(gè)分支上,但其他基因型蟲(chóng)株的聚類沒(méi)有規(guī)律。蛋白二級(jí)結(jié)構(gòu)和抗原表位預(yù)測(cè)結(jié)果表明,GRA14蛋白經(jīng)過(guò)3次跨膜,主要由6個(gè)α-螺旋、3個(gè)β折疊、11個(gè)β轉(zhuǎn)角和若干個(gè)無(wú)規(guī)則卷曲構(gòu)成,并預(yù)測(cè)有4個(gè)B細(xì)胞抗原表位。表明GRA14蛋白可以作為抗弓形蟲(chóng)病疫苗候選分子研制弓形蟲(chóng)病亞單位疫苗及表位疫苗。
[Abstract]:In order to study the genetic variation of Toxoplasma gondii dense granule protein 14 (GRA14) gene, 9 strains of Toxoplasma gondii DNA from different hosts and geographical sources were used as templates. The GRA14 gene of Toxoplasma gondii (Toxoplasma gondii) was amplified by PCR, and the PCR product was cloned into pMD18-T vector and sequenced. Genetic variation of GRA14 gene was analyzed by bioinformatics software and phylogenetic tree was constructed. The structure of GRA14 protein was analyzed and antigen epitopes were predicted with antigen index. The results showed that the length of GRA14 gene of 9 strains of Toxoplasma gondii was 1 227bp. the A T content of the sequence was between 46.94 and 47.43%, and the nucleotide variation rate of the corresponding sequence of Toxoplasma gondii ME49 strain was between 0.08% and 0.73%.
