【摘要】：目的研制狂犬病毒(RV)核蛋白(NP)時間分辨免疫熒光分析(TRFIA)試劑盒。方法采用雙抗體夾心法建立RV NP TRFIA試劑盒,對試劑盒的各項指標(biāo)進行評價。結(jié)果試劑盒準(zhǔn)確度好,線性范圍為5～2 500mEU/mL,靈敏度為1.018mEU/mL,精密度良好,分析內(nèi)精密度為2.7%～9.3%,分析間精密度為3.5%～12.2%。將15份疫苗樣品用本法與國內(nèi)常用ELISA檢測試劑盒同時檢測,其相關(guān)系數(shù)r為0.85,穩(wěn)定性試驗表明試劑可以在4℃穩(wěn)定半年,37℃穩(wěn)定7d。結(jié)論試劑盒各項已檢測指標(biāo)達(dá)到臨床檢驗要求,有望進一步作臨床檢測試驗。
[Abstract]:Objective to develop a time resolved immunofluorescence assay (TRFIA) kit for nucleoprotein NPV of rabies virus (RV). Methods the RV NP TRFIA kit was established by double antibody sandwich method, and the indexes of the kit were evaluated. The calibration curve was linear in the range of 5 ~ 2 500 mEU / mL, with a sensitivity of 1.018 mEU-mL. the precision was good, the analytical precision was 2. 7% and 9. 3%, and the analytical precision was 3. 5% and 12. 2%. 15 samples of vaccine were detected by this method and the ELISA kit used in our country. The correlation coefficient r is 0.85. The stability test shows that the reagent can be stabilized at 4 鈩，
