高濃度氧氣吸入對(duì)ARDS大鼠肺組織氧化應(yīng)激的影響

【摘要】:目的探討ARDS大鼠吸入高濃度氧氣后肺組織氧化應(yīng)激的變化。方法實(shí)驗(yàn)動(dòng)物分為正常大鼠組和ARDS大鼠組,每組30只,每組再以吸入空氣、60%氧氣、100%氧氣各分成三個(gè)亞組,每個(gè)亞組10只。72 h后留取動(dòng)脈血行血氧分壓(PO2)及丙二醛(MDA)、超氧化物歧化酶(SOD)、過(guò)氧化氫酶(CAT)、單胺氧化酶(MAO)測(cè)定;肺組織行HE染色觀察Smith評(píng)分,RT-PCR檢測(cè)Ⅱ型肺泡上皮細(xì)胞(AECⅡ)特異性肺泡表面活性蛋白C(SPC)和Ⅰ型肺泡上皮細(xì)胞(AECⅠ)特異性水通道蛋白5(AQP5)的mRNA表達(dá)。結(jié)果各組大鼠吸氧72 h后均存活。與正常大鼠空氣亞組比較,正常大鼠100%氧氣亞組Smith評(píng)分升高(3.77±1.20比0.83±0.26),P0.05;MDA升高(2.39±0.92比0.82±0.31),P0.05;SOD下降(51.99±21.73比86.44±26.38),P0.05;AQP5 mRNA下降(0.67±0.25比0.91±0.28),P0.05;ARDS大鼠空氣亞組的Smith評(píng)分升高(11.65±3.82比0.83±0.26),P0.05;MDA升高(5.73±2.43比0.82±0.31),P0.05;SOD下降(45.71±14.68比86.44±26.38),P0.05;CAT下降(0.52±0.23比0.85±0.34),P0.05;AQP5 mRNA下降(0.28±0.12比0.91±0.28),P0.05;SPC mRNA下降(0.33±0.14比0.72±0.28),P0.05。ARDS組大鼠空氣亞組的Smith評(píng)分(11.65±3.82比3.77±1.20),P0.05和AQP5 mRNA(0.28±0.12比0.67±0.25),P0.05;與正常大鼠100%氧氣亞組差異顯著。MAO在各組內(nèi)和組間均差異無(wú)顯著性(P0.05)。ARDS組大鼠動(dòng)脈血氧分壓100%氧氣亞組高于60%氧氣亞組(112.86±26.32比86.94±21.07),P0.05,60%氧氣亞組高于空氣亞組(86.94±21.07比65.05±15.19),P0.05。在ARDS組內(nèi)與空氣亞組比較,60%氧氣亞組(0.43±0.18 VS 0.28±0.12),P0.05和100%氧氣亞組(0.47±0.23比0.28±0.12),P0.05的AQP5 mRNA均有顯著性升高;100%氧氣亞組的Smith評(píng)分降低(7.88±2.53比11.65±3.82),P0.05,SPC mRNA升高(0.58±0.25比0.33±0.14),P0.05。結(jié)論高濃度氧氣吸入和ARDS均可以影響氧自由基,氧自由基并不是高濃度氧的特征性產(chǎn)物,難以作為客觀有效的氧毒性監(jiān)測(cè)指標(biāo);高濃度氧氣吸入對(duì)成年大鼠的肺部損害輕微且沒(méi)有特異性;ARDS大鼠吸入100%氧氣后沒(méi)有出現(xiàn)疊加損害,反而比60%氧氣更好地改善了肺部Smith評(píng)分和動(dòng)脈血氧分壓,增加了SPCmRNA和AQP5mRNA的表達(dá),提示高濃度氧氣對(duì)ARDS可能存在更為復(fù)雜的修復(fù)機(jī)制。
[Abstract]:Objective to investigate the changes of oxidative stress in lung tissue of ARDS rats after inhaling high concentration of oxygen. Methods the experimental animals were divided into normal rat group and ARDS rat group with 30 rats in each group. Each group was divided into three subgroups with inhaled air, 60% oxygen and 100% oxygen, 10 rats in each subgroup. 72 hours later, arterial blood was taken for partial pressure of oxygen (PO2) and malondialdehyde (MDA),) (SOD), catalase (CAT), monoamine oxidase (MAO).
