油酸致急性肺損傷乳豬模型的建立

【摘要】:目的: 采用油酸溶于等容無水酒精混合豬血清持續(xù)勻速注射方法,構(gòu)建符合肺損傷標(biāo)準(zhǔn)、血流動力學(xué)穩(wěn)定的乳豬急性肺損傷動物模型。 方法: 采用改良法構(gòu)建油酸型乳豬急性肺損傷模型:選用高純度油酸,以每公斤體重0.1ml的劑量溶于等容無水酒精,然后與10ml自制豬血清經(jīng)渦旋振蕩器充分混勻后,再用微泵注射器經(jīng)乳豬頸靜脈插管在15min內(nèi)緩慢勻速的注入。出生25-30天的乳豬12只,隨機分為兩組:a)實驗組(Study group,n=6):油酸組,用上述方法注射油酸。b)對照組(Control group,n=6):生理鹽水組。用上述方法注射等量生理鹽水。注射過程中檢測Pa02,當(dāng)Pa02/FiO2比值300mmHg,即認為模型建立成功。監(jiān)測肺功能指標(biāo)如動脈血氧分壓、動脈血二氧化碳分壓,PH值及血流動力學(xué)指標(biāo)如心率、平均動脈血壓、心輸出量、中心靜脈壓;實驗結(jié)束,取右下肺組織標(biāo)本,生理鹽水沖洗標(biāo)本,甲醛固定標(biāo)本,放于容器中,行病理檢查。了解其組織學(xué)改變情況。 結(jié)果: 對照組肺功能指標(biāo)動脈血氧分壓、動脈血二氧化碳分壓、pH值及血流動力學(xué)指標(biāo)心率、平均動脈壓、心輸出量、中心靜脈壓基礎(chǔ)值無顯著性差異(P0.05)。持續(xù)勻速注射油酸完成后,實驗組很快即達到急性肺損傷模型要求。Pa02/Fi02比值下降明顯,平均動脈血壓、心率、心輸出量、動脈氧分壓、pH值與對照組相比明顯降低(P0.05);實驗組動脈二氧化碳分壓、心率比對照組明顯升高(P0.05)。病理組織學(xué)切片顯示實驗組病變較對照組明顯嚴(yán)重。 結(jié)論:1.通過油酸混合豬血清持續(xù)勻速注射方法,可以構(gòu)建血流動力學(xué)穩(wěn)定的乳豬肺損傷模型; 2.通過油酸混合豬血清持續(xù)勻速注射方法,能夠反映臨床典型的急性肺損傷/急性呼吸窘迫綜合癥(ALI/ARDS)病理表現(xiàn);以這種模型為基礎(chǔ),可以深入開展ALI/ARDS相關(guān)課題的研究。
[Abstract]:Aim: to establish an animal model of acute lung injury in suckling pigs with stable hemodynamics according to lung injury standard by continuous uniform injection of oleic acid in serum of isovolumetric anhydrous alcohol mixed pigs. Methods: the acute lung injury model of oleic acid type suckling pig was established by modified method. High purity oleic acid was selected and dissolved in isovolumetric anhydrous alcohol at the dose of 0.1ml per kilogram of body weight, and then mixed with 10ml pig serum by vortex oscillator. Then micropump syringe was used to infuse the jugular vein of the suckling pig slowly and uniformly into 15min. Twelve suckling pigs born 25-30 days were randomly divided into two groups: control group (n = 6): study group (n = 6): oleic acid group (n = 6): control group (n = 6): normal saline group (n = 6). The same amount of saline was injected with the above method. Pa02 was detected during injection, and when the ratio of Pa02 / FiO2 was 300 mmHg, the model was considered successful. Pulmonary function parameters such as arterial partial pressure of oxygen, arterial blood pressure of carbon dioxide and PH value and hemodynamic indexes such as heart rate, mean arterial blood pressure, cardiac output, central venous pressure were monitored. The specimens were washed with normal saline, fixed by formaldehyde and placed in containers for pathological examination. Learn about the histological changes. Results: in the control group, there was no significant difference in arterial oxygen pressure, arterial carbon dioxide pressure and pH value, heart rate, mean arterial pressure, cardiac output and central venous pressure (P0.05). After continuous and uniform injection of oleic acid, the experimental group reached the requirement of acute lung injury model quickly. The ratio of Pa02 / Fi02 decreased significantly, the mean arterial blood pressure, heart rate, cardiac output, arterial oxygen pressure and pH value decreased significantly compared with the control group (P 0.05). The arterial carbon dioxide partial pressure and heart rate in the experimental group were significantly higher than those in the control group (P 0.05). Histopathological sections showed that the pathological changes in the experimental group were significantly more serious than those in the control group. Conclusion 1. The hemodynamic stable lung injury model of suckling pig can be established by continuous uniform injection of oleic acid mixed pig serum. 2. Continuous uniform injection of oleic acid mixed pig serum can reflect the typical clinical manifestations of acute lung injury / acute respiratory distress syndrome (Ali / ARDS), and based on this model, the research on ALI / ARDS related research can be carried out in depth.
