芹菜素逆轉(zhuǎn)白介素6誘導(dǎo)的中性粒細(xì)胞凋亡延遲

【摘要】:目的:觀察芹菜素對(duì)炎性因子白細(xì)胞介素6(interleakin-6,IL-6)刺激的中性粒細(xì)胞(neutrophils,Neu)凋亡的影響,并探討可能的分子機(jī)制。方法:采取健康志愿者外周靜脈血,分離純化Neu,隨機(jī)分為對(duì)照組、芹菜素組、IL-6組、IL-6+芹菜素組及加入廣譜天冬氨酸特異性半胱氨酸蛋白酶(caspase)抑制劑的Q-VD-Oph+IL-6+芹菜素組、z-VAD-fmk+IL-6+芹菜素組。流式細(xì)胞術(shù)觀察Neu凋亡率,Western blot檢測(cè)Neu活化的caspase-3(cleaved caspase-3)的表達(dá)情況。結(jié)果:與對(duì)照組比較,芹菜素促進(jìn)Neu凋亡(P0.05),IL-6抑制Neu凋亡(P0.05);兩者共孵育時(shí),芹菜素可以部分逆轉(zhuǎn)IL-6誘導(dǎo)的Neu凋亡延遲(P0.05);與IL-6組比較,芹菜素與IL-6共孵育組cleaved caspase-3的蛋白表達(dá)水平顯著增高(P0.05),而且芹菜素對(duì)caspase-3的活化作用和對(duì)IL-6誘導(dǎo)的Neu凋亡延遲的逆轉(zhuǎn)作用可以被caspase抑制劑消除(P0.05)。結(jié)論:芹菜素可能通過caspase途徑逆轉(zhuǎn)IL-6誘導(dǎo)的Neu凋亡延遲,為今后治療Neu炎癥性氣道疾病提供了新思路。
[Abstract]:Aim: to investigate the effect of apigenin on neutrophil (neutrophils Neu) apoptosis induced by interleukin 6 (IL 6) interleakin-6 (IL 6) and to explore the possible molecular mechanism. Methods: Neu was isolated and purified from peripheral venous blood of healthy volunteers. It was randomly divided into control group, IL-6 group and Q-VD-Oph IL-6 apigenin group added with broad-spectrum aspartate specific cysteine protease caspase inhibitor, z-VAD-fmk IL-6 apigenin group. The apoptosis rate of Neu and the expression of caspase-3(cleaved caspase-3 activated by Neu were detected by flow cytometry and Western blot. Results: compared with the control group, apigenin promoted the apoptosis of Neu and inhibited the apoptosis of Neu by P0.05IL-6. When incubated with apigenin, the apoptosis of Neu induced by IL-6 was partially reversed by apigenin, and compared with that of IL-6 group. The expression level of cleaved caspase-3 protein was significantly increased in cleaved caspase-3 co-incubated with apigenin and IL-6. Moreover, the activation of caspase-3 by apigenin and the reversal of Neu apoptosis induced by IL-6 could be eliminated by caspase inhibitor. Conclusion: apigenin may reverse the delayed apoptosis of Neu induced by IL-6 via caspase pathway and provide a new idea for the treatment of Neu inflammatory airway disease.
