17β-雌二醇對(duì)小鼠肺成纖維細(xì)胞細(xì)胞周期的影響

【摘要】:目的:觀察17β-雌二醇(17β-E2)對(duì)小鼠肺成纖維細(xì)胞細(xì)胞周期的影響。方法:根據(jù)L9(33)正交實(shí)驗(yàn)設(shè)計(jì),用不同質(zhì)量濃度SiO2(20、50、100 mg/L)誘導(dǎo)小鼠肺成纖維細(xì)胞,再用不同濃度17β-E2(10-6、10-7、10-8mol/L)干預(yù)不同時(shí)間(24、48、72 h),MTT法檢測小鼠肺成纖維細(xì)胞的增殖狀況,以增殖活性較低為篩選標(biāo)準(zhǔn),選取最佳實(shí)驗(yàn)條件。將小鼠肺成纖維細(xì)胞分成3組:空白對(duì)照組、SiO2誘導(dǎo)組和17β-E2干預(yù)組,在相應(yīng)實(shí)驗(yàn)條件下培養(yǎng)細(xì)胞,運(yùn)用流式細(xì)胞術(shù)檢測細(xì)胞周期。結(jié)果:最佳實(shí)驗(yàn)條件篩選結(jié)果為:用20 mg/L SiO2誘導(dǎo),用10-6mol/L 17β-E2干預(yù)24 h。3組G1期細(xì)胞比例、S+G2期細(xì)胞比例差異均有統(tǒng)計(jì)學(xué)意義(F=50.300、29.896,P0.001)。SiO2誘導(dǎo)組G1期細(xì)胞比例較空白對(duì)照組降低,S+G2期細(xì)胞比例升高;而17β-E2干預(yù)組G1期細(xì)胞比例較SiO2誘導(dǎo)組升高,S+G2期細(xì)胞比例降低(P0.05)。結(jié)論:17β-E2可能是通過阻滯小鼠肺成纖維細(xì)胞由G1期進(jìn)入S期和G2期,從而抑制其增殖。
[Abstract]:Aim: to observe the effect of 17 尾 -estradiol 17 尾 -E _ 2 on the cell cycle of mouse lung fibroblasts. Methods: mouse lung fibroblasts were induced by different concentrations of SiO2O2050mg / L) according to the L9 / 33) orthogonal design. The proliferation of mouse lung fibroblasts was detected by MTT assay with different concentrations of 17 尾 -E2O10-6 (10-7) 10-8 mol / L at different time points. The optimum experimental conditions were selected with low proliferative activity as the screening criterion. The mouse lung fibroblasts were divided into three groups: the blank control group induced by SiO2 and the 17 尾 -E2 intervention group. The cells were cultured under the corresponding experimental conditions, and the cell cycle was detected by flow cytometry. Results: the optimal experimental conditions were as follows: induced by 20 mg/L SiO2, The ratio of G 1 phase cells and S G 2 phase cells in 10-6mol/L 17 尾 -E2 group were significantly different from those in the control group. The ratio of G 1 phase cells in the G 1 phase group induced by 10-6mol/L 17 尾 -E2 was significantly lower than that in the control group, and the percentage of S G 2 phase cells in the S G 2 phase group was significantly higher than that in the control group. The proportion of G 1 phase cells in 17 尾 -E2 treated group was higher than that in SiO2 induced group, and the proportion of S G 2 phase cells was decreased (P 0.05). Conclusion the proliferation of mouse lung fibroblasts may be inhibited by blocking the proliferation of mouse lung fibroblasts from G _ 1 to S and G _ 2.
