篩選自人卵巢癌細(xì)胞系HO8910的干細(xì)胞生物學(xué)特性分析

【摘要】:目的探討篩選自人卵巢癌細(xì)胞系HO8910干細(xì)胞的生物學(xué)特性。方法將前期篩選卵巢癌細(xì)胞系HO8910的干細(xì)胞在無血清培養(yǎng)基中進(jìn)行傳代培養(yǎng),以HO8910細(xì)胞作為對照。體外觀察兩種細(xì)胞的球體形成能力;將兩種細(xì)胞接種于含血清培養(yǎng)基,觀察其分化能力,并檢測干細(xì)胞相關(guān)標(biāo)記物;采用藥敏試驗(yàn)檢測兩種細(xì)胞對順鉑、多柔比星及米托蒽醌的敏感性;將無血清培養(yǎng)基中進(jìn)行傳代培養(yǎng)的兩種細(xì)胞接種于NOD/SCID裸鼠,觀察其體內(nèi)成瘤情況。結(jié)果 HO8910細(xì)胞在無血清培養(yǎng)基中的球體形成能力明顯低于卵巢癌細(xì)胞(P0.05)。卵巢癌干細(xì)胞分化前CD133、CD117陽性表達(dá)率明顯高于分化后(P均0.05)。卵巢癌干細(xì)胞分化后ABCG2、Nanog、Oct4、BCRP、E-cadherin表達(dá)均低于分化前(P均0.05),與HO8910細(xì)胞分化前后上述指標(biāo)比較差異均無統(tǒng)計(jì)學(xué)意義(P均0.05)。0.25、0.5μg/m L順鉑,0.5、1.5μmol/L多柔比星,0.05、0.25μg/m L米托蒽醌作用于卵巢癌干細(xì)胞、HO8910細(xì)胞48 h時(shí),卵巢癌干細(xì)胞的相對活性均高于HO8910細(xì)胞(P均0.05)。裸鼠接種1×103個(gè)卵巢癌干細(xì)胞即可成瘤;隨著干細(xì)胞接種數(shù)量的增加,成瘤比率逐漸增加,成瘤時(shí)間逐漸縮短(P均0.05)。結(jié)論篩選自人卵巢癌細(xì)胞系HO8910的卵巢癌干細(xì)胞具有自我更新和分化、體內(nèi)成瘤、高表達(dá)干細(xì)胞基因以及對化療耐藥等特性,符合腫瘤干細(xì)胞的理論標(biāo)準(zhǔn)。
[Abstract]:Objective to investigate the biological characteristics of human ovarian cancer cell line HO8910. Methods the stem cells of ovarian cancer cell line HO8910 were subcultured in serum-free medium and HO8910 cells were used as control. The spherical formation ability of the two kinds of cells was observed in vitro, the differentiation ability of the two kinds of cells was observed, the differentiation ability was observed, and the related markers of stem cells were detected. The sensitivity of the two kinds of cells to DDP, doxorubicin and mitoxantrone was detected by drug sensitivity test. The two kinds of cells subcultured in serum-free medium were inoculated into NOD/SCID nude mice to observe their tumorigenesis in vivo. Results the globular formation ability of HO8910 cells in serum-free medium was significantly lower than that of ovarian cancer cells (P 0.05). The positive expression rate of CD133,CD117 in ovarian cancer stem cells before differentiation was significantly higher than that after differentiation. The expression of ABCG2,Nanog,Oct4,BCRP,E-cadherin in ovarian cancer stem cells after differentiation was lower than that before differentiation, but there was no significant difference compared with that before and after differentiation of HO8910 cells. 0.25 渭 g / mL DDP, 0.5 渭 mol / L doxorubicin, 0.05 渭 g / mL mitoxantrone and 0.25 渭 g / mL mitoxantrone on ovarian cancer stem cells for 48 h. The relative activity of ovarian cancer stem cells was higher than that of HO8910 cells. With the increase of the number of stem cells inoculated with 1 脳 103 ovarian cancer stem cells in nude mice, the tumorigenesis rate increased gradually and the tumorigenesis time decreased gradually (P 0.05). Conclusion the ovarian cancer stem cells screened from human ovarian cancer cell line HO8910 have the characteristics of self-renewal and differentiation, tumorigenesis in vivo, high expression of stem cell genes and resistance to chemotherapy, which is in accordance with the theoretical standard of tumor stem cells.
