妊娠期糖尿病患者胎盤(pán)組織胰島素受體底物1、蛋白酪氨酸磷酸酶1B表達(dá)及意義

【摘要】:目的探討妊娠期糖尿病(GDM)患者胎盤(pán)組織胰島素受體底物1(IRS-1)、蛋白酪氨酸磷酸酶1B(PTP1B)表達(dá)及意義。方法選擇GDM孕婦49例(GDM組),正常足月妊娠孕婦50例(正常組)。兩組均于孕24~28周檢測(cè)空腹胰島素(FINS)、空腹血糖(FPG),采用胰島素穩(wěn)態(tài)模型評(píng)估法(HOMA)計(jì)算胰島素抵抗指數(shù)(HOMAIR)、胰島β細(xì)胞功能(HOMA-β%)、胰島素敏感指數(shù)(HOMA-ISI);兩組分娩后取胎盤(pán)組織,采用免疫組化ABC法檢測(cè)胎盤(pán)組織IRS-1、PTP 1B。結(jié)果 GDM組FPG、FINS、HOMA-IR均高于正常組(P均0.01),HOMA-ISI、HOMA-β%均低于正常組(P均0.01)。正常組胎盤(pán)組織IRS-1陽(yáng)性表達(dá)率為100%,GDM組為63.26%,兩組比較P0.01。正常組胎盤(pán)組織PTP 1B陽(yáng)性表達(dá)率為56.00%,GDM組為91.83%,兩組比較P0.01。GDM患者胎盤(pán)組織PTP 1B與IRS-1表達(dá)呈負(fù)相關(guān)(r=-4.85,P0.01);IRS-1與HOMA-IR呈負(fù)相關(guān)(r=-0.552,P0.01),與HOMA-β%和HOMA-ISI無(wú)相關(guān)性(P均0.05);PTP 1B與HOMA-IR呈正相關(guān)(r=0.563,P0.01),與HOMA-β%和HOMA-ISI無(wú)相關(guān)性(P均0.05)。結(jié)論 GDM患者胎盤(pán)組織IRS-1表達(dá)降低,PTP 1B表達(dá)升高,且均與HOMA-IR相關(guān);胰島素信號(hào)傳導(dǎo)系統(tǒng)中信號(hào)蛋白表達(dá)異?赡芘cGDM發(fā)病有關(guān)。
[Abstract]:Objective to investigate the expression and significance of insulin receptor substrate 1 (IRS-1) and protein tyrosine phosphatase 1B (PTP1B) in placenta of patients with gestational diabetes mellitus (GDM). Methods 49 GDM pregnant women (GDM group) and 50 normal term pregnant women (normal group) were selected. Fasting insulin (FINS), fasting blood glucose (FPG), was measured at 24 脳 28 weeks of gestation. Insulin homeostasis model (HOMA) was used to calculate insulin resistance index (HOMAIR), islet 尾 cell function (HOMA- 尾%) and insulin sensitivity index (HOMA-ISI). Placental tissues were taken after delivery and placental tissue IRS-1,PTP 1B was detected by ABC method. Results FPG,FINS,HOMA-IR in GDM group was higher than that in normal group (P < 0.01), and HOMA-ISI,HOMA- 尾% was lower than that in normal group (P < 0.01). The positive expression rate of IRS-1 in placenta was 100% in normal group and 63.26% in GDM group.
