脂肪酸結(jié)合蛋白4 DNA甲基化在L-NAME干預胎盤滋養(yǎng)細胞脂代謝異常中的作用

【摘要】:目的觀察并分析一氧化氮合酶抑制劑L-NAME對滋養(yǎng)細胞脂肪酸結(jié)合蛋白4(FABP4)基因啟動子區(qū)DNA甲基化及脂質(zhì)代謝異常的影響。方法體外培養(yǎng)人胎盤滋養(yǎng)細胞(HTR-8)并用100μmol/L-NAME干預48 h后檢測胎盤滋養(yǎng)細胞脂質(zhì)含量;qRT-PCR和Western blot檢測胎盤滋養(yǎng)細胞中DNA甲基轉(zhuǎn)移酶1(DNMT1)、FABP4 m RNA和蛋白表達改變;巢式甲基化特異性PCR(NT-MSP)檢測胎盤滋養(yǎng)細胞中FABP4啟動子區(qū)DNA甲基化的程度改變。結(jié)果 L-NAME干預滋養(yǎng)細胞后脂質(zhì)含量明顯增多,且細胞內(nèi)FABP4 mRNA及蛋白表達顯著增加(P0.05),而FABP4基因啟動子區(qū)甲基化程度以及DNMT1的表達水平顯著減低。結(jié)論 FABP4啟動子區(qū)低DNA甲基化可能參與了L-NAME誘導的胎盤滋養(yǎng)細胞脂代謝異常的調(diào)控過程。
[Abstract]:Objective to observe and analyze the effects of nitric oxide synthase inhibitor L-NAME on DNA methylation and lipid metabolism in the promoter region of trophoblast fatty acid binding protein 4 (FABP4) gene. Methods Human placental trophoblasts (HTR-8) were cultured in vitro and treated with 100 渭 mol / L-NAME for 48 h. The expression of DNA methyltransferase 1 (DNMT1), FABP4 m RNA and protein in placental trophoblast was detected by qRT-PCR and Western blot, and the degree of DNA methylation in FABP4 promoter region in placental trophoblast was detected by nest methylated specific PCR (NT-MSP). Results after L-NAME intervention, the lipid content was significantly increased, and the expression of FABP4 mRNA and protein in the cells was significantly increased (P 0.05), while the degree of methylation in the promoter region of FABP4 gene and the expression of DNMT1 were significantly decreased. Conclusion low DNA methylation in FABP4 promoter region may be involved in the regulation of abnormal lipid metabolism in placental trophoblast induced by L-NAME.
