辣椒堿對人宮頸癌細胞Hela和C-33A的抗腫瘤活性

【摘要】:目的:研究辣椒堿對Hela(HPV+)和C-33A(HPV-)細胞生長增殖和凋亡的影響,探討辣椒堿對宮頸癌細胞的抑制作用與相關機制。方法:選取HPV(+)的Hela和HPV(-)的C-33A宮頸癌細胞株為研究對象。CCK-8法檢測細胞活性,流式細胞術檢測辣椒堿對細胞生長的抑制作用、細胞周期和對細胞凋亡的影響;q PCR和Western blot法檢測細胞凋亡相關基因m RNA和蛋白表達。結果:辣椒堿對宮頸癌Hela和C-33A細胞增殖抑制呈濃度和時間依賴性;辣椒堿能顯著促進兩種細胞凋亡,對Hela細胞的促凋亡效果更明顯。辣椒堿處理后,兩種細胞的細胞色素C(cytchrome C)和caspase3表達量顯著增加,Bax表達量增加,Bcl-2表達量降低。結論:辣椒堿可顯著抑制宮頸癌Hela和C-33A細胞增殖和促進細胞的凋亡,其機理可能是通過誘導細胞內(nèi)活性氧類物質(zhì)(ROS)增加、細胞色素C和Caspase3表達量增加等途徑,最終誘導細胞凋亡。
