干性基因Oct-4和SOX-2在宮頸癌中的表達

【摘要】:目的: 研究干性基因Oct-4和SOX-2在宮頸癌中的表達情況,探討這二個基因的表達水平能否成為判斷宮頸癌愈后的重要指標。 材料與方法: (1)蛋白水平研究:52例宮頸癌標本均取自2008-2012年間吉林大學第一醫(yī)院婦產(chǎn)科手術(shù)治療患者,病理學檢查確診宮頸癌,所有病例術(shù)前均無化療、放療和免疫治療病史。28例正常宮頸組織標本來源于因子宮肌瘤或其他良性疾病行全子宮切除術(shù)患者,全部患者無相關(guān)癌癥病史。通過免疫組織化學SP法檢測干性基因Oct-4和SOX-2在宮頸癌和正常宮頸組織中的表達。(2)基因水平研究:選取行宮頸癌根治術(shù)患者的新鮮宮頸癌組織12例,,所有病例術(shù)前均無化療、放療和免疫治療病史;選取因子宮良性疾病行全子宮切除術(shù)患者的新鮮正常宮頸組織6例。通過Q-PCR的方法,檢測宮頸癌中Oct-4mRNA和SOX-2mRNA的表達差異。 結(jié)果: (1)免疫組織化學染色結(jié)果顯示:①Oct-4在宮頸癌組織中呈陽性表達,其表達率為46.1%(24/52),顯著高于在正常組織中的7.1%(2/28),組間比較差異有統(tǒng)計學意義(p0.005);②SOX-2在宮頸癌組織中呈陽性表達,其表達率為59.6%(31/52),顯著高于在正常組織中的10.7%(3/28),組間比較差異有統(tǒng)計學意義(p 0.005);③宮頸癌組織中Oct-4和SOX-2的表達與患者年齡、病理類型和FIGO分期均無關(guān)聯(lián)性;與宮頸癌的分化程度有關(guān),隨分化程度的降低Oct-4和SOX-2的表達水平升高,其組間比較差異均有統(tǒng)計學意義,呈負相關(guān);④宮頸癌組織中,Oct-4與SOX-2蛋白的表達具有正相關(guān)性關(guān)系(p 0.05)。(2)Q-PCR實驗結(jié)果顯示:①SOX-2mRNA在宮頸癌組織中的表達情況(12.67+3.98)高于其在正常宮頸組織的表達情況(1.5+0.548),差異具有統(tǒng)計學意義(t=9.53,p<0.01);②Oct-4mRNA在宮頸癌組織中的表達情況(15.83+3.24)高于其在正常宮頸組織的表達情況(1.333+0.516),差異具有統(tǒng)計學意義(t=15.11,p<0.01);③兩者表達未見明顯正相關(guān)性。 結(jié)論: 1在蛋白和mRNA水平上,干性基因Oct-4在宮頸癌組織中表達量均高于正常宮頸組織。 2在蛋白和mRNA水平上,干性基因SOX-2在宮頸癌組織中表達量均高于正常宮頸組織。 3干性基因Oct-4和SOX-2的表達水平可能成為判斷宮頸癌愈后的潛在指標。
[Abstract]:Objective: to study the expression of dry gene Oct-4 and SOX-2 in cervical cancer and to explore whether the expression level of these two genes can be an important index to judge the prognosis of cervical cancer. Materials and methods: (1) protein level study: 52 cases of cervical cancer were collected from the patients with gynecology and obstetrics surgery in the first Hospital of Jilin University from 2008 to 2012. The history of radiotherapy and immunotherapy. 28 cases of normal cervical tissues were collected from patients undergoing hysterectomy due to uterine leiomyoma or other benign diseases. The expression of dry genes Oct-4 and SOX-2 in cervical cancer and normal cervix tissues was detected by immunohistochemical SP method. (2) Gene level: 12 cases of fresh cervical cancer tissues with radical cervical cancer resection were selected. All cases had no history of chemotherapy, radiotherapy and immunotherapy before operation. The fresh normal cervical tissues of 6 patients with benign uterine diseases undergoing total hysterectomy were selected. The expression of Oct-4mRNA and SOX-2mRNA in cervical cancer was detected by Q-PCR. Results: (1) Immunohistochemical staining showed that the positive expression rate of 1Oct-4 in cervical carcinoma was 46.1% (24 / 52), which was significantly higher than that in normal tissues (7.1%, 2 / 28). The difference between the two groups was statistically significant (p 0.005). The positive expression rate of 2SOX-2 in cervical cancer tissues was 59.6% (31 / 52), which was significantly higher than that in normal tissues (10.7%, 3 / 28). The difference between the two groups was statistically significant (p 0.005). 3There was no correlation between the expression of Oct-4 and SOX-2 in cervical carcinoma and age, pathological type and FIGO stage. With the decrease of the differentiation degree, the expression of Oct-4 and SOX-2 increased, and the difference between the two groups was statistically significant and negatively correlated with the degree of differentiation. (4) in the tissues of cervical cancer, There was a positive correlation between the expression of Oct-4 and SOX-2 protein (p0.05). (2). The results of Q-PCR assay showed that the expression of 1SOX-2mRNA in cervical cancer tissues (12.67 3.98) was higher than that in normal tissues. Cervical tissue expression (1.5 0.548), The difference was statistically significant (t = 9.53, p < 0.01). The expression of 2Oct-4mRNA in cervical carcinoma (15.83 3.24) was higher than that in normal cervix (1.333 0.516). 3There was no significant positive correlation between the two expressions. Conclusion: 1 at the level of protein and mRNA, the expression of dry gene Oct-4 in cervical carcinoma is higher than that in normal cervix. 2 at the level of protein and mRNA, the expression of dry gene SOX-2 in cervical cancer was higher than that in normal cervix. 3 the expression level of dry gene Oct-4 and SOX-2 may be a potential index to judge the prognosis of cervical cancer.
