苦參堿及聯(lián)合順鉑對(duì)宮頸癌SiHa細(xì)胞中TSLC1基因表達(dá)的影響

【摘要】:目的探討苦參堿及聯(lián)合順鉑對(duì)宮頸癌Si Ha細(xì)胞中TSLC1基因表達(dá)的影響。方法以人宮頸癌細(xì)胞株Si Ha細(xì)胞株為試驗(yàn)材料,采取甲基偶氮唑藍(lán)法檢測(cè)并計(jì)算細(xì)胞抑制率。采取實(shí)時(shí)熒光逆轉(zhuǎn)錄聚合酶聯(lián)反應(yīng)法分析TSLC1 mRNA基因表達(dá)情況。結(jié)果順鉑、苦參堿聯(lián)合順鉑均可顯著抑制Si Ha細(xì)胞體外增殖,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);隨著用藥濃度的增加,抑制作用增強(qiáng),不同濃度兩兩比較,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);聯(lián)合組細(xì)胞抑制率高于苦參組、順鉑組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。三組用藥后,TSLC1基因表達(dá)水平均高于對(duì)照組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);隨著濃度的增加,影響效果更強(qiáng),組間不同濃度兩兩比較,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);聯(lián)合組高于順鉑組、苦參堿組,同濃度比較,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);順鉑組與苦參堿組同濃度比較,差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論苦參堿及聯(lián)合順鉑對(duì)宮頸癌Si Ha細(xì)胞中TSLC1基因表達(dá)有抑制作用,且與藥物濃度呈劑量依賴性。
[Abstract]:Objective to investigate the effect of matrine and cisplatin on the expression of TSLC1 gene in cervical cancer Si Ha cells. Methods Human cervical cancer cell line Si Ha cell line was used as the experimental material, and the inhibition rate was measured by methyl azolium blue method. The expression of TSLC1 mRNA gene was analyzed by real-time fluorescence reverse transcriptase polymerase chain reaction (RT-PCR). Results Cisplatin, matrine and cisplatin could significantly inhibit the proliferation of Si Ha cells in vitro (P0.05). The difference was statistically significant (P0.05); the cell inhibition rate of combined group was higher than that of Sophora flavescens group, cisplatin group, the difference was statistically significant (P0.05). The expression level of TSLC1 gene in the three groups was significantly higher than that in the control group (P0.05); with the increase of the concentration, the effect was stronger, the difference was statistically significant (P0.05); the combined group was higher than the cisplatin group (P0.05). Matrine group, the same concentration comparison, the difference was statistically significant (P0.05); cisplatin group and matrine group with the same concentration comparison, the difference was not statistically significant (P0.05). Conclusion Matrine and cisplatin can inhibit the expression of TSLC1 gene in cervical cancer Si Ha cells in a dose-dependent manner.
