【摘要】：目的探討母體高鹽飲食對(duì)子代腎臟血管發(fā)育的影響及其相應(yīng)機(jī)制。方法自然懷孕的Sprague-Dawley大鼠隨機(jī)分為高鹽組和對(duì)照組,高鹽組孕鼠懷孕期間給予含8%NaCl的高鹽飼料,對(duì)照組給予含1%NaCl的正常飼料,兩組孕鼠均正常飲水。子代出生后,所有母鼠恢復(fù)正常飼料,新生鼠正常哺乳至1個(gè)月分籠。取成年子代公鼠腎臟葉間動(dòng)脈進(jìn)行血管功能檢測(cè)、單個(gè)血管平滑肌(vascular smooth muscle cell,VSMC)細(xì)胞電生理實(shí)驗(yàn)。結(jié)果高鹽組子代腎臟小動(dòng)脈對(duì)去氧腎上腺素(phenylephrine,Phe)引起的收縮反應(yīng)強(qiáng)于對(duì)照組(P0.05),蛋白激酶C(protein kinase C,PKC)非選擇性阻斷劑GF109203X對(duì)Phe收縮反應(yīng)的作用兩組間差異也有統(tǒng)計(jì)學(xué)意義(P0.05);電生理實(shí)驗(yàn)結(jié)果顯示:Phe在兩組子代腎臟小血管VSMC中,均可抑制大電導(dǎo)鈣激活性鉀通道(high-conductance Ca~(2+)-activated K+channel,BK通道)電流,但在高鹽組的作用更為明顯(P0.05),GF109203X的存在使BK電流在Phe作用前后差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論孕期高鹽飲食可能通過(guò)改變Phe介導(dǎo)的成年♂性子代腎臟小動(dòng)脈血管收縮的敏感性、PKC對(duì)BK通道的調(diào)控作用,從而影響其腎臟血管功能的發(fā)育,并可能由此增加其子代腎臟疾病發(fā)生的易感性。
[Abstract]:Objective to investigate the effect of maternal high salt diet on renal vascular development in offspring and its mechanism. Methods the naturally pregnant Sprague-Dawley rats were randomly divided into high salt group and control group. The pregnant rats in high salt group were given high salt diet containing 8%NaCl during pregnancy, while the control group were given normal diet containing 1%NaCl. After the birth of the offspring, all the female rats returned to normal feed, and the newborn mice suckled to 1 month cage. The vascular function of renal interlobar artery of adult male rat was measured and the electrophysiological experiment of single vascular smooth muscle (vascular smooth muscle cell,VSMC) cells was carried out. Results the contractile response of renal arterioles to noradrenaline (phenylephrine,Phe) in high salt group was stronger than that in control group (P0.05). The effect of GF109203X, a non-selective inhibitor of protein kinase C (protein kinase, on Phe contraction was significantly different between the two groups (P0.05). The results of electrophysiological experiments showed that in two groups of small renal vessel VSMC, The high-conductance Ca~ (2) -activated K channel,BK channel (high-conductance Ca~ (2) -activated K channel,BK channel) current was inhibited, but the effect was more obvious in high salt group (P0.05). The existence of GF109203X resulted in no significant difference in BK current before and after Phe treatment (P0.05). Conclusion High salt diet during pregnancy may affect the development of renal vascular function by altering the sensitivity of Phe mediated renal arterioles contraction and the regulation of BK channels. This may increase the susceptibility to kidney disease in their offspring.
