【摘要】：目的:探討核轉(zhuǎn)錄因子5(NFAT5)在重度子癇前期(sPE)胎盤(pán)中的表達(dá)及其與圍生兒結(jié)局的相關(guān)性。方法:收集2015年1月至2016年1月在鄭州大學(xué)第三附屬醫(yī)院行剖宮產(chǎn)分娩的sPE孕婦(試驗(yàn)組)60例。選取同期非PE行剖宮產(chǎn)術(shù)的孕婦60例作為對(duì)照組。免疫組化法檢測(cè)胎盤(pán)組織中NFAT5表達(dá);RT-PCR法檢測(cè)胎盤(pán)組織中NFAT5 mRNA表達(dá)。對(duì)胎盤(pán)中NFAT5 mRNA水平與圍生兒結(jié)局進(jìn)行相關(guān)分析。結(jié)果:sPE組的新生兒體重、胎盤(pán)重量、Apgar評(píng)分及分娩孕周均低于對(duì)照組(P均0.05)。兩組胎盤(pán)中NFAT5蛋白多位于胎盤(pán)滋養(yǎng)細(xì)胞的胞核與胞質(zhì)中,絨毛間質(zhì)細(xì)胞內(nèi)有少量表達(dá)。sPE組的NFAT5蛋白和mRNA表達(dá)水平均高于對(duì)照組,差異均有統(tǒng)計(jì)學(xué)意義(t=-40.434,P=0.001;t=-6.696,P=0.001)。sPE組的NFAT5 mRNA相對(duì)表達(dá)量與新生兒出生體重、胎盤(pán)重量、分娩孕周均呈負(fù)相關(guān)(r分別為-0.554,-0.698,-0.612,P均0.05),與孕婦體重指數(shù)(BMI)呈正相關(guān)(r=0.630,P=0.012)。結(jié)論:sPE組胎盤(pán)組織中NFAT5表達(dá)水平升高,與圍生兒不良結(jié)局有關(guān)。
[Abstract]:Aim: to investigate the expression of nuclear transcription factor 5 (NFAT5) in placenta of severe preeclampsia (sPE) and its correlation with perinatal outcome. Methods: 60 sPE pregnant women (trial group) who were delivered by cesarean section in the third affiliated Hospital of Zhengzhou University from January 2015 to January 2016 were collected. Sixty pregnant women with non-PE cesarean section were selected as control group. The expression of NFAT5 in placenta was detected by immunohistochemistry. RT-PCR was used to detect the expression of NFAT5 mRNA in placenta. The correlation between placental NFAT5 mRNA level and perinatal outcome was analyzed. Results the newborns weight, placental weight Apgar score and gestational week of delivery were significantly lower in the control group than in the control group (all P 0.05). The NFAT5 protein in placenta of both groups was mostly located in the nucleus and cytoplasm of placental trophoblast, and the expression of NFAT5 protein and mRNA in the villus interstitial cells was higher than that in the control group. The relative expression of NFAT5 mRNA was negatively correlated with birth weight, placental weight and gestational week of delivery (r = -0.554-0.698U -0.612P0.05, respectively), and positively correlated with (BMI) (r0.630P0.012). Conclusion the increased expression of NFAT5 in placental tissue of SPE group is related to perinatal adverse outcome.
