DAPT抑制妊娠滋養(yǎng)細胞腫瘤Notch信號通路對細胞增殖和EMT影響的研究

【摘要】:目的:探討Notch信號通路在人妊娠滋養(yǎng)細胞腫瘤發(fā)生發(fā)展過程中的重要作用,及其與EMT之間的關系。方法:采用γ-分泌酶抑制劑DAPT處理JAR和JEG-3兩種人絨毛膜癌細胞株,MTT法檢測兩種細胞株的增殖情況,q PCR法檢測Notch1 mRNA表達。Western blot法檢測DAPT阻斷Notch信號通路后JAR細胞中EMT相關蛋白E-cad的表達情況。結果:DAPT能抑制JEG-3、JAR細胞生長,10μmol/L和15μmol/L DAPT作用48h開始分別抑制JEG-3和JAR細胞生長(P0.05),且抑制作用呈時間和濃度依賴關系。JEG-3細胞中,1μmol/L和10μmol/L DAPT作用組中Notch1 mRNA表達下調,兩組比較差異無統(tǒng)計學意義(P0.05);JAR細胞中,10μmol/L和15μmol/L DAPT作用組中Notch1 mRNA表達下調,兩組比較差異有統(tǒng)計學意義(P0.05)。DAPT能上調JAR細胞中E-cad表達。結論:Notch信號通路和EMT與妊娠滋養(yǎng)細胞腫瘤的發(fā)生發(fā)展有關;γ-分泌酶抑制劑DAPT或可成為妊娠滋養(yǎng)細胞腫瘤治療的一個新靶點。
[Abstract]:Aim: to investigate the important role of Notch signaling pathway in the development of human gestational trophoblastic tumor and its relationship with EMT. Methods: the proliferation of two kinds of human choriocarcinoma cell lines (JAR and JEG-3) was detected by DAPT, a 緯 -secretory enzyme inhibitor. The expression of Notch1 mRNA was detected by q-PCR and EMT in JAR cells after DAPT blocked Notch signaling pathway was detected by Western blot. Expression of related protein E-cad. Results the growth of JEG-3,JAR cells was inhibited by 10 渭 mol/L and 15 渭 mol/L DAPT for 48 h, respectively (P0.05). The inhibitory effect was in a time-and concentration-dependent manner. Notch1 mRNA expression was down-regulated in JEG-3 cells treated with 1 渭 mol/L and 10 渭 mol/L DAPT, respectively. There was no significant difference between the two groups (P0.05) the expression of Notch1 mRNA was down-regulated in the group treated with 10 渭 mol/L and 15 渭 mol/L DAPT in jar cells, and the difference between the two groups was statistically significant (P0.05) .DAPT could up-regulate the expression of E-cad in JAR cells. Conclusion the EMT and the signal pathway of Notch are related to the development of gestational trophoblastic tumors, and 緯 -secretase inhibitor DAPT may be a new target for the treatment of gestational trophoblastic tumors.
