【摘要】：目的:探討細(xì)胞命運(yùn)決定因子DACH1(dachshund family transcription factor 1)對(duì)人子宮內(nèi)膜癌HEC-1A細(xì)胞體外增殖能力和細(xì)胞周期的影響。方法 :將DACH1過(guò)表達(dá)的重組慢病毒LV-DACH1-GFP感染人子宮內(nèi)膜癌HEC-1A細(xì)胞,在激光共聚焦顯微鏡下觀察DACH1在HEC-1A細(xì)胞中的定位;分別采用實(shí)時(shí)熒光定量PCR法和蛋白質(zhì)印跡法檢測(cè)HEC-1A細(xì)胞中DACH1 m RNA和蛋白的表達(dá),MTT法檢測(cè)HEC-1A細(xì)胞的體外的增殖能力,FCM法檢測(cè)細(xì)胞周期的分布。結(jié)果 :重組慢病毒LV-DACH1-GFP感染人子宮內(nèi)膜癌HEC-1A細(xì)胞后,LV-DACH1-GFP主要分布在細(xì)胞核中。LV-DACH1-GFP感染組HEC-1A細(xì)胞中,DACH1 m RNA和蛋白的表達(dá)水平均明顯高于對(duì)照組(HEC-1A細(xì)胞感染慢病毒LV-GFP)和空白組(HEC-1A細(xì)胞未進(jìn)行任何感染)(P值均0.01),LV-DACH1-GFP感染組HEC-1A細(xì)胞的增殖受到明顯抑制(P0.05),細(xì)胞周期阻滯于G0/G1期(P0.01)。結(jié)論 :D ACH1基因可抑制人子宮內(nèi)膜癌HEC-1A細(xì)胞的增殖,這一作用可能與DACH1基因阻滯細(xì)胞周期有關(guān)。
[Abstract]:Aim: to investigate the effect of DACH1 (dachshund family transcription factor 1 on the proliferation and cell cycle of human endometrial carcinoma HEC-1A cells in vitro. Methods: human endometrial carcinoma HEC-1A cells were infected with recombinant lentivirus LV-DACH1-GFP which was overexpressed by DACH1. The localization of DACH1 in HEC-1A cells was observed under confocal laser microscope. The expression of DACH1 m RNA and protein in HEC-1A cells were detected by real-time fluorescence quantitative PCR assay and Western blotting respectively. The proliferation of HEC-1A cells in vitro was detected by FCM method and the cell cycle distribution was detected by FCM method. Results: after recombinant lentivirus LV-DACH1-GFP was infected with human endometrial carcinoma HEC-1A cells, the expression levels of RNA and protein in HEC-1A cells of LV-DACH1-GFP infection group were significantly higher than those of control group (HEC-1A cells infected with lentivirus LV-GFP) and empty cells (LV-DACH1-GFP). The proliferation of LV-DACH1-GFP infected HEC-1A cells was significantly inhibited (P0.05), and the cell cycle was blocked in G0/G1 phase (P0.01). ConclusionThe cell proliferation of human endometrial carcinoma HEC-1A cells can be inhibited by DACH1 D ACH1 gene, which may be related to the arrest of cell cycle by DACH1 gene.
