上皮性卵巢癌中FAT4和YAP1的表達及意義

【摘要】:目的:卵巢上皮性腫瘤是女性生殖系統(tǒng)最常見的卵巢腫瘤,而且卵巢癌一經(jīng)確診,超過70%已是中晚期,從而導(dǎo)致了容易復(fù)發(fā)、預(yù)后差以及病死率高等特點,嚴(yán)重威脅了女性的生命和生活質(zhì)量。因此國內(nèi)外學(xué)者在惡性腫瘤的發(fā)生和發(fā)展機制中做了非常多的研究,其中上皮組織的Hippo信號通路備受關(guān)注,目前多數(shù)學(xué)者研究證實,惡性腫瘤的發(fā)生發(fā)展與Hippo信號通路調(diào)控出現(xiàn)異常有著密切的關(guān)系,主要是促進組織細胞的增殖與抑制細胞的凋亡,加強腫瘤細胞的侵襲和遷移能力,導(dǎo)致腫瘤形成及遠處轉(zhuǎn)移。其中FAT4參與Hippo信號通路的調(diào)控,YAP1是Hippo信號通路下游的十分重要的分子,參與著惡性腫瘤的發(fā)生與發(fā)展。本研究采用免疫組織化學(xué)方法檢測FAT4及Yes-相關(guān)蛋白1(YAP1)在上皮性卵巢癌中的表達情況,并對其臨床意義及兩者在卵巢癌中的相關(guān)性進行分析,從而為卵巢癌的研究提供參考和依據(jù)。方法1、研究對象:收集我院2014年12月至2015年12月期間初治的卵巢上皮性癌組織標(biāo)本45例(其中漿液性卵巢癌31例,粘液性卵巢癌14例)。收集同期手術(shù)并經(jīng)病理證實的卵巢上皮性良性腫瘤組織標(biāo)本25例(其中漿液性囊腺瘤18例,粘液性囊腺瘤7例)依照MDACC組織分級標(biāo)準(zhǔn)進行統(tǒng)計研究。2、標(biāo)本采集:采集術(shù)后卵巢腫瘤組織,大小約1*1*1cm,生理鹽水沖洗凈后,至-80℃冰箱冷凍保存?zhèn)溆?后進行石蠟包埋等處理。3檢測指標(biāo)及方法:應(yīng)用免疫組織化學(xué)方法檢測兩組組織標(biāo)本中FAT4、YAP1的表達情況,分析其與上皮性卵巢癌的臨床病理分級及FAT4與YAP1之間的相關(guān)性。4數(shù)據(jù)分析:采用統(tǒng)計軟件SPSS16.0對實驗結(jié)果進行統(tǒng)計學(xué)分析,兩組間率的比較采用卡方檢驗,采用Spearman秩相關(guān)分析方法,分析FAT4和YAP1之間的相互關(guān)系。取?=0.05作為檢驗水準(zhǔn),P0.05為有統(tǒng)計學(xué)差異。結(jié)果:(1)FAT4及YAP1在上皮性卵巢癌細胞表達率分別為84.4%和75.5%,而在卵巢上皮良性腫瘤中表達率分別為44%和40%,有統(tǒng)計學(xué)差異(P0.05)。本研究發(fā)現(xiàn)FAT4和YAP1的表達率在與卵巢癌的分化程度相關(guān)(P0.05)(2)應(yīng)用Spearman對FAT4與YAP1表達的相關(guān)性進行相關(guān)分析,顯示FAT4與YAP1在上皮性卵巢癌表達呈正相關(guān)(r=0.565,P0.001)。結(jié)論:FAT4及YAP1蛋白在上皮性卵巢癌中的表達明顯升高,與卵巢癌的病理分級相關(guān)。通過本研究,我們認為FAT4和YAP1可以作為診斷上皮性卵巢癌的生物指標(biāo),可作為上皮性卵巢癌組織分級判斷的參考指標(biāo)?梢詾槁殉舶┑陌邢蛑委熖峁┮环N新思路。
[Abstract]:Objective: ovarian epithelial tumor is the most common ovarian tumor in female reproductive system. A serious threat to women's lives and quality of life. Therefore, scholars at home and abroad have done a lot of research on the pathogenesis of malignant tumor, and the Hippo signaling pathway of epithelial tissue has attracted much attention. The occurrence and development of malignant tumor is closely related to the abnormal regulation of Hippo signaling pathway, which mainly promotes the proliferation of hippo cells, inhibits cell apoptosis, strengthens the ability of invasion and migration of tumor cells, and leads to tumor formation and distant metastasis. FAT4 is involved in the regulation of Hippo signaling pathway and YAP1 is a very important molecule downstream of Hippo signaling pathway, which is involved in the occurrence and development of malignant tumor. In this study, the expression of FAT4 and Yes-associated protein 1 (YAP1) in epithelial ovarian carcinoma was detected by immunohistochemical method, and the clinical significance and the correlation between FAT4 and YAS-associated protein 1 in ovarian cancer were analyzed. So as to provide a reference and basis for the study of ovarian cancer. Methods 1. Participants: 45 cases of epithelial ovarian carcinoma (including 31 cases of serous ovarian cancer and 14 cases of mucinous ovarian cancer) were collected from our hospital from December 2014 to December 2015. A total of 25 cases of ovarian epithelial benign tumors, including 18 cases of serous cystadenoma, were collected. 7 cases of mucinous cystadenoma) were statistically studied according to MDACC tissue grading standard.
