【摘要】：目的觀察宮頸癌組織中血管生成素-2(Ang-2)和其受體酪氨酸激酶受體2(Tie-2)表達(dá)變化,并探討其臨床意義。方法選擇2014年1月~2017年1月收治的86例宮頸癌患者的手術(shù)切除宮頸癌組織標(biāo)本為觀察組,同期收治的86例子宮肌瘤患者的正常宮頸組織標(biāo)本為對(duì)照組。采用免疫組化法定性檢測(cè)兩組Ang-2、Tie-2蛋白表達(dá)情況。采用RT-PCR法定量檢測(cè)兩組Ang-2、Tie-2 mRNA表達(dá)量,采用Western blotting法定量檢測(cè)兩組Ang-2、Tie-2蛋白表達(dá)量。分析宮頸癌組織Ang-2、Tie-2蛋白定性表達(dá)情況與宮頸癌臨床病理參數(shù)的關(guān)系。結(jié)果觀察組Ang-2、Tie-2 mRNA及蛋白表達(dá)量均高于對(duì)照組(P均0.05)。免疫組化染色結(jié)果觀察組Ang-2表達(dá)陽性51例(59.30%)、Tie-2表達(dá)陽性54例(62.79%),對(duì)照組Ang-2表達(dá)陽性35例(40.70%)、Tie-2表達(dá)陽性37例(43.02%),觀察組Ang-2、Tie-2蛋白陽性表達(dá)率均高于對(duì)照組(P均0.05)。宮頸癌組織Ang-2、Tie-2陽性表達(dá)與宮頸癌FIGO分期、淋巴結(jié)轉(zhuǎn)移有關(guān)(P均0.05),與年齡、組織學(xué)類型、分化程度無關(guān)(P均0.05)。結(jié)論宮頸癌組織Ang-2和其受體Tie-2表達(dá)上調(diào)。檢測(cè)宮頸癌組織Ang-2和其受體Tie-2表達(dá)有助于判斷宮頸癌患者的FIGO分期和淋巴結(jié)轉(zhuǎn)移情況。
[Abstract]:Objective to observe the expression of angiopoietin-2 Ang-2) and its receptor tyrosine kinase receptor 2-Tie-2 in cervical carcinoma and to explore its clinical significance. Methods 86 cases of cervical carcinoma treated from January 2014 to January 2017 were selected as observation group and 86 cases of normal cervical tissue as control group. The expression of Ang-2 tie-2 protein was detected qualitatively by immunohistochemical method. The expression of Ang-2 tie-2 mRNA in two groups was detected by RT-PCR assay, and the expression of Ang-2 tie-2 protein was detected by Western blotting assay. To analyze the relationship between the qualitative expression of Ang-2 tie-2 protein and the clinicopathological parameters of cervical carcinoma. Results the expression of mRNA and protein of Ang-2 tie-2 in the observation group was higher than that in the control group (P 0.05). The results of immunohistochemical staining showed that there were 51 cases of positive expression of Ang-2 in observation group and 54 cases of positive expression of Tie-2 in 59.30 cases, while 35 cases of positive expression of Ang-2 in control group were positive in 37 cases (43.022%). The positive rate of Ang-2 tie-2 protein in observation group was higher than that in control group (P 0.05). The positive expression of Ang-2pTie-2 in cervical carcinoma was correlated with FIGO stage, lymph node metastasis (P < 0.05), and had no correlation with age, histological type and differentiation degree (P < 0.05). Conclusion the expression of Ang-2 and its receptor Tie-2 is up-regulated in cervical carcinoma. The expression of Ang-2 and its receptor Tie-2 in cervical carcinoma is helpful to judge the FIGO stage and lymph node metastasis.
