TWEAK通過外泌體途徑介導巨噬細胞抑制上皮性卵巢癌細胞轉移的研究

【摘要】:目的:探討TWEAK能否通過外泌體途徑介導巨噬細胞對上皮性卵巢癌細胞轉移的調控。方法:佛波酯誘導人單核細胞系THP-1為貼壁的巨噬細胞,TWEAK刺激巨噬細胞24h后抽提外泌體,透射電鏡觀察其形態(tài);分別將PBS、巨噬細胞分泌的外泌體、TWEAK刺激后巨噬細胞分泌的外泌體與上皮性卵巢癌細胞系SKOV3、HO-8910pm共培養(yǎng),48h后通過Transwell實驗檢測其遷移、侵襲能力。結果:透射電鏡可見巨噬細胞分泌的外泌體呈圓形,直徑30~100nm。Transwell實驗顯示,與空白對照組相比,巨噬細胞分泌的外泌體可使上皮性卵巢癌細胞的遷移和侵襲能力增高;而TWEAK刺激巨噬細胞后,其分泌的外泌體則可降低其遷移和侵襲能力。結論:TWEAK可通過外泌體途徑逆轉巨噬細胞對上皮性卵巢癌細胞的促轉移作用。
[Abstract]:Aim: to investigate whether TWEAK mediates macrophages' regulation of epithelial ovarian cancer cell metastasis through exocrine pathway. Methods: phorbol ester induced human monocytes THP-1 as adherent macrophages to stimulate macrophages for 24 hours, then extract exocrine bodies, and observe their morphology by transmission electron microscope (TEM). PBSs, macrophages secreted by TWEAK and epithelial ovarian cancer cell line SKOV3HO-8910pm were co-cultured for 48 hours. The migration and invasion ability were detected by Transwell assay. Results: the exocrine bodies secreted by macrophages were round by transmission electron microscope. The diameter 30~100nm.Transwell test showed that the exocrine secreted by macrophages could increase the migration and invasion ability of epithelial ovarian cancer cells compared with the blank control group. However, after TWEAK stimulated macrophages, the exocrine secreted by macrophages decreased their migration and invasion ability. Conclusion: TWEAK can reverse the role of macrophages in promoting metastasis of epithelial ovarian cancer cells through exocrine pathway.
