【摘要】：目的:探討葉酸對(duì)宮頸癌Si Ha細(xì)胞及He La細(xì)胞自噬相關(guān)蛋白p62表達(dá)的影響。方法:在正常葉酸水平(正常對(duì)照組,1.0μg/ml)和低劑量葉酸水平(低劑量葉酸組,0.1μg/ml)兩種條件下分別干預(yù)宮頸癌Si Ha細(xì)胞及He La細(xì)胞,并于24小時(shí)、48小時(shí)、72小時(shí)分別收取細(xì)胞,采用實(shí)時(shí)熒光定量PCR實(shí)驗(yàn)及免疫印跡實(shí)驗(yàn)分別檢測(cè)自噬相關(guān)蛋白p62 m RNA水平及蛋白水平表達(dá)量的變化。結(jié)果:1.與正常對(duì)照組相比,低劑量葉酸組培養(yǎng)的宮頸癌Si Ha細(xì)胞,在蛋白水平其48小時(shí)、72小時(shí)的p62的表達(dá)量降低(P0.05),在m RNA水平僅72小時(shí)的p62表達(dá)量降低(P0.05)。2.與正常對(duì)照組相比,低劑量葉酸組培養(yǎng)的宮頸癌He La細(xì)胞,無(wú)論在蛋白水平還是m RNA水平,僅48小時(shí)的p62表達(dá)量降低(P0.05)。結(jié)論:1.低劑量葉酸使宮頸癌Si Ha細(xì)胞自噬相關(guān)蛋白p62的表達(dá)下降,提示低劑量葉酸可誘導(dǎo)宮頸癌細(xì)胞自噬的發(fā)生。2.低劑量葉酸使宮頸癌He La細(xì)胞自噬相關(guān)蛋白p62的表達(dá)下降,同樣提示低劑量葉酸可誘導(dǎo)宮頸癌細(xì)胞自噬的發(fā)生。
[Abstract]:Aim: to investigate the effect of folic acid on the expression of autophagy associated protein p62 in Cervical carcinoma SIHA cells and He-La cells.Methods: normal folic acid (normal control group, 1.0 渭 g / ml) and low-dose folic acid (low dose folic acid group, 0.1 渭 g / ml) were used to treat cervical cancer cell line SIHA and He-La cells, respectively. Cells were collected at 24 hours, 48 hours and 72 hours, respectively.The level and expression of autophagy associated protein p62 m RNA were detected by real-time fluorescence quantitative PCR assay and Western blotting assay respectively.The result is 1: 1.Compared with the normal control group, the expression of p62 in the low dose folic acid group was decreased at the protein level at 48 hours and 72 hours, and the expression of p62 at m RNA level at 72 hours was decreased.Compared with the normal control group, the expression of p62 in He-La cells cultured with low dose folic acid at the protein level and m RNA level decreased only 48 hours.Conclusion 1.Low dose folic acid decreased the expression of autophagy associated protein p62 in Cervical carcinoma SIHA cells, suggesting that low dose folic acid could induce the occurrence of autophagy in cervical cancer cells.Low dose folic acid decreased the expression of autophagy associated protein p62 in helium La cells, suggesting that low dose folic acid could induce autophagy in cervical cancer cells.
