不明原因早期復發(fā)性流產患者蛻膜自然殺傷細胞對滋養(yǎng)層細胞系侵襲功能的影響

【摘要】:目的研究早孕期復發(fā)性流產(RSA)患者與正常早孕婦女的蛻膜自然殺傷細胞(d NK)對滋養(yǎng)層細胞系HTR-8/SVneo侵襲能力的影響。方法收集正常早孕人工流產婦女(正常早孕組,n=10)和不明原因RSA患者(RSA組,n=8)的早孕蛻膜組織,用密度梯度離心法分離出蛻膜淋巴細胞,免疫磁珠法進一步分選出CD56+CD16-NK細胞,即蛻膜NK細胞,將蛻膜NK細胞與HTR-8/SVneo共培養(yǎng)24 h后,通過MTT法和Transwell侵襲實驗檢測蛻膜NK細胞對滋養(yǎng)層細胞系HTR8/SVneo黏附及侵襲能力的影響,用Real-time PCR檢測共培養(yǎng)后HTR-8/SVneo細胞的MMP-2和MMP-9 mRNA水平的表達情況。結果與正常早孕婦女相比,不明原因早孕期RSA患者的蛻膜NK細胞對滋養(yǎng)層細胞系HTR8/SVneo侵襲能力有減弱作用,并使HTR-8/SVneo表達促侵襲分子MMP-2和MMP-9表達降低。結論妊娠早期局部母-胎界面d NK細胞的功能異?赡苁菍е虏幻髟騌SA的一個原因。
[Abstract]:Objective to study the effect of decidual natural killer (NK) on the invasion of trophoblastic cell line HTR-8/SVneo in patients with recurrent early pregnancy (RSA) and normal early pregnant women.Methods the decidual tissues of women with normal early pregnancy induced abortion (normal early pregnancy group) and unexplained RSA group were collected. Decidual lymphocytes were isolated by density gradient centrifugation, and CD56 CD16-NK cells were further isolated by immunomagnetic beads.Decidual NK cells were co-cultured with HTR-8/SVneo for 24 h. The effects of decidual NK cells on the adhesion and invasion of trophoblastic cell line HTR8/SVneo were detected by MTT and Transwell invasion assay.The expression of MMP-2 and MMP-9 mRNA in co-cultured HTR-8/SVneo cells was detected by Real-time PCR.Results compared with normal early pregnant women, decidua NK cells of unexplained early pregnancy RSA patients decreased the invasiveness of trophoblastic cell line HTR8/SVneo, and decreased the expression of MMP-2 and MMP-9 in HTR-8/SVneo.Conclusion the abnormal function of d NK cells at the maternal-fetal interface in early pregnancy may be one of the causes of unexplained RSA.
