【摘要】：目的： 檢測激活素（Activin）受體在胎兒生長受限胎盤及正常足月胎兒胎盤中的定位及表達水平，研究其與胎兒生長受限的關(guān)系，探討激活素受體在胎兒生長受限發(fā)病機制中的作用。 方法： 各收集15例足月胎兒生長受限和正常足月妊娠孕婦分娩后的胎盤組織。HE染色法分析各組胎盤組織；免疫組織化學(xué)法分析各組胎盤組織中激活素受體的表達部位，并進行半定量分析；Real-Time PCR法測定各組胎盤組織中激活素受體基因mRNA的表達。 結(jié)果： 1、HE染色結(jié)果：對照組胎盤組織絨毛發(fā)育良好，，絨毛血管擴張明顯，間質(zhì)細(xì)胞少，絨毛滋養(yǎng)細(xì)胞以合體滋養(yǎng)細(xì)胞為主；FGR組胎盤組織絨毛發(fā)育欠佳，絨毛血管擴張不良，絨毛內(nèi)間質(zhì)增生，鈣化、壞死增多。 2、免疫組化染色結(jié)果：兩組胎盤組織中激活素受體均主要表達于合體滋養(yǎng)細(xì)胞及細(xì)胞滋養(yǎng)細(xì)胞胞漿中。FGR組胎盤組織中激活素受體ⅠA、ⅠB染色陽性程度與對照組相比，差異無統(tǒng)計學(xué)意義（P＞0.05）；激活素受體ⅡA、ⅡB染色陽性程度均顯著高于對照組，差異有統(tǒng)計學(xué)意義（P＜0.05）。 3、Real-Time PCR結(jié)果：FGR組胎盤組織中激活素受體ⅠA、Ⅰ B基因表達與對照組相比，差異無統(tǒng)計學(xué)意義(P＞0.05)；激活素受體Ⅱ A、 ⅡB基因表達均較對照組升高，差異有統(tǒng)計學(xué)意義(P＜0.05)。 結(jié)論： 1、胎兒生長受限患者胎盤組織中激活素受體ⅠA、Ⅰ B表達水平與正常妊娠相比無差異，激活素受體ⅡA、Ⅱ B表達水平較正常妊娠明顯升高。 2、妊娠晚期胎盤組織中激活素受體ⅡA、 ⅡB表達水平的上升可能是導(dǎo)致胎兒生長受限發(fā)生的一個重要因素。 3、激活素及其受體共同參與調(diào)節(jié)胎兒宮內(nèi)生長發(fā)育，激活素受體的紊亂可能在胎兒生長受限的發(fā)病機制中發(fā)揮重要的作用。
[Abstract]:Objective:. To detect the localization and expression of Activinin receptor in placenta of fetal growth restriction and normal term fetal placenta, to study the relationship between activin receptor and fetal growth restriction, and to explore the role of activin receptor in the pathogenesis of fetal growth restriction. Methods:. The placental tissues of 15 pregnant women with full-term fetal growth restriction and normal term pregnancy were collected. The placenta tissues were analyzed by HE staining, and the expression sites of activin receptors in placenta tissues were analyzed by immunohistochemical method. The expression of activin receptor gene (mRNA) in placenta was determined by semi-quantitative analysis with Real-Time PCR. Results:. 1the results of HE staining showed that the placental villi of the control group developed well, the villus vasodilated obviously, the interstitial cells were few, the villus trophoblast was mainly composed of syncytiotrophoblast and the villi of the FGR group were not well developed and the villi were dilated poorly. Interstitial hyperplasia, calcification and necrosis in villi. 2. Immunohistochemical staining: Activin receptors were mainly expressed in the cytoplasm of syncytiotrophoblast and cytotrophoblast.
