4.1N蛋白抑制人卵巢透明細(xì)胞癌細(xì)胞遷移侵襲及其機(jī)制初探

【摘要】:目的:探討4.1N蛋白對卵巢透明細(xì)胞癌細(xì)胞遷移和侵襲能力的調(diào)控并初步探討相關(guān)分子機(jī)制。方法:應(yīng)用免疫組化法檢測4.1N蛋白在卵巢透明細(xì)胞癌組織中的表達(dá)缺失狀況。建立4.1N穩(wěn)定轉(zhuǎn)染卵巢透明細(xì)胞癌ES-2細(xì)胞系,通過劃痕實(shí)驗(yàn)、Transwell遷移和侵襲實(shí)驗(yàn)觀察細(xì)胞遷移侵襲能力的改變。利用qRT-PCR和Western blot法檢測緊密連接相關(guān)分子(Claudin-4和ZO-1)、上皮-間質(zhì)轉(zhuǎn)化(EMT)標(biāo)記物(E-cadherin、Ncadherin和Vimentin)和相關(guān)轉(zhuǎn)錄因子(Twist、Slug和ZEB-2)及基質(zhì)金屬蛋白酶(MMP-2和MMP-3)表達(dá)水平的變化。結(jié)果:與卵巢異位的子宮內(nèi)膜組織相比,卵巢透明細(xì)胞癌中4.1N蛋白表達(dá)水平顯著降低甚至缺失(P0.0001)。過表達(dá)4.1N后,ES-2細(xì)胞的遷移和侵襲能力顯著降低,Claudin-4和ZO-1表達(dá)上調(diào),Twist、Slug、ZEB-2、MMP-2和MMP-3表達(dá)水平下降。結(jié)論:4.1N在卵巢透明細(xì)胞癌細(xì)胞中可能通過調(diào)控緊密連接蛋白、部分上皮-間質(zhì)轉(zhuǎn)化轉(zhuǎn)錄因子和基質(zhì)金屬蛋白酶的表達(dá)水平,抑制癌細(xì)胞的遷移和侵襲能力,進(jìn)而參與負(fù)性調(diào)控卵巢透明細(xì)胞癌的演進(jìn)。
[Abstract]:Objective: to investigate the regulation of 4.1 N protein on the migration and invasion of ovarian clear cell carcinoma cells and to explore the molecular mechanism. The expression of 4.1N protein in clear cell carcinoma of ovary was detected by immunohistochemical method. The ES-2 cell line of ovarian clear cell carcinoma was established by 4.1N stable transfection. Through the scratch test. Transwell migration and invasion assay were used to observe the change of cell migration and invasion ability. QRT-PCR and Western blot were used to detect the tightly linked molecules (. Claudin-4 and ZO-1). E-cadherin and Vimentin (E-cadherin and Vimentin) and related transcription factor Twist. Slug and ZEB-2) and matrix metalloproteinase (MMP-2) and MMP-3) expression levels. Results: compared with ovarian ectopic endometrium tissue. The expression level of 4.1N protein in ovarian clear cell carcinoma was significantly decreased or even deleted P0.0001N, and the migration and invasion ability of Ovarian clear cell carcinoma was significantly decreased after the expression of 4.1 N. The expression of Claudin-4 and ZO-1 up-regulated the expression of Twistus Slugfus ZEB-2. Conclusion the expression of MMP-2 and MMP-3 in ovarian clear cell carcinoma cells may be regulated by tight junction protein. The expression of transcriptional factors and matrix metalloproteinases (MMPs) in some epithelium-mesenchymal cells inhibits the migration and invasion of cancer cells and thus participates in the negative regulation of the progression of ovarian clear cell carcinoma.
