基于核磁共振的800m跑訓(xùn)練前后尿液代謝組學(xué)特征

【摘要】:背景:目前研究發(fā)現(xiàn)800 m跑的代謝特征不是十分明晰,為800 m的運(yùn)動(dòng)訓(xùn)練造成了很大的阻礙。目的:了解800 m跑項(xiàng)目的物質(zhì)和能量代謝特征,提高800 m跑訓(xùn)練的科學(xué)化水平,驗(yàn)證基于核磁共振的尿液代謝組學(xué)技術(shù)在運(yùn)動(dòng)訓(xùn)練監(jiān)測(cè)方面的可行性。方法:對(duì)某高校運(yùn)動(dòng)訓(xùn)練專(zhuān)業(yè)7名中跑2級(jí)運(yùn)動(dòng)員進(jìn)行一次極限強(qiáng)度800 m跑,結(jié)合人體代謝數(shù)據(jù)庫(kù)(HMDB)等數(shù)據(jù)庫(kù),對(duì)運(yùn)動(dòng)前、后的尿液進(jìn)行代謝組學(xué)相關(guān)分析。結(jié)果與結(jié)論:OPLS-DA模型顯示運(yùn)動(dòng)前和運(yùn)動(dòng)后樣本的代謝模式呈現(xiàn)簇類(lèi)分布。造成這一變化的特征性代謝物主要有亮氨酸、乳酸、肌酐、琥珀酸、次黃嘌呤、腺苷等代謝物。通過(guò)分析可以推斷出800 m跑是以糖酵解供能為主、磷酸原供能和有氧氧化供能共同參與的運(yùn)動(dòng)項(xiàng)目,800 m跑后運(yùn)動(dòng)員氨基酸代謝、甲烷代謝等代謝過(guò)程較為旺盛,而且在800 m跑的過(guò)程中會(huì)出現(xiàn)一定程度的氧化應(yīng)激。結(jié)果說(shuō)明基于核磁共振的尿液代謝組學(xué)技術(shù)有無(wú)創(chuàng)性、靈敏度高、樣品需求量少、處理簡(jiǎn)單等優(yōu)勢(shì)。
[Abstract]:Background: at present, it is found that the metabolic characteristics of 800m running are not very clear, which is a great obstacle for 800m sports training. Objective: to understand the characteristics of substance and energy metabolism in 800m running events, improve the scientific level of 800m running training, and verify the feasibility of urine metabonomics technology based on nuclear magnetic resonance (NMR) in monitoring exercise training. Methods: seven middle and second grade athletes in a certain university were run with the limit strength of 800 m, and the urine before and after exercise was analyzed with the database of Human body Metabolism Database (HMDB) and other databases. Results and conclusions the OPLS-DA model showed that the metabolic patterns of pre-exercise and post-exercise samples showed cluster distribution. The main metabolites responsible for this change are leucine, lactic acid, creatinine, succinic acid, Hypoxanthine, adenosine and other metabolites. It can be inferred from the analysis that the main metabolic process of 800 m running is glycolysis, the metabolism of amino acid, methane and so on after 800 m running. And there will be a certain degree of oxidative stress during the 800 m run. The results showed that the urinary metabolomics technique based on nuclear magnetic resonance had the advantages of noninvasive, high sensitivity, less sample demand and simple treatment.
