2010-2011年恩施和武漢柯薩奇病毒A16型VP1基因特征分析

【摘要】:目的了解恩施和武漢地區(qū)手足口病(HFMD)的分子流行病學特征和病原體柯薩奇病毒A16型(CA16)VP1的基因特征。方法收集2010年4月至2011年8月來自于恩施和武漢地區(qū)的189例HFMD患兒咽拭子標本,接種病毒易感Vero細胞,對產(chǎn)生明顯細胞病變效應的標本進行初篩和PCR鑒定;對陽性毒株VP1基因序列進行擴增,構建pMD18-T-VP1重組克隆,然后測定其VP1全長序列,通過MEGA5軟件構建系統(tǒng)進化樹進行系統(tǒng)進化分析,通過DNAstar軟件對VP1保守表位的氨基酸序列保守性進行分析。結果手足口病患者以≤5歲嬰幼兒為主,男性多于女性。從189例手足口病患兒咽拭子標本中分離到7株CA16,VP1序列進化分析顯示均屬于B1亞型。對VP1基因上的6個抗原表位進行分析,其中的3個區(qū)域呈現(xiàn)高度保守性,這些保守區(qū)可被用于疫苗研究。結論 CA16是2010-2011年恩施和武漢地區(qū)手足口病的重要病原體之一,屬于B1基因型。
[Abstract]:Objective to investigate the molecular epidemiological characteristics of HFMD and the gene characteristics of Coxsackie virus A16 (CA16) VP1 in Enshi and Wuhan. Methods from April 2010 to August 2011, the throat swabs of 189 patients with HFMD from Enshi and Wuhan were collected and inoculated with virulent Vero cells. The specimens with obvious cytopathic effect were screened and identified by PCR. The VP1 gene sequence of the positive strain was amplified, the recombinant pMD18-T-VP1 clone was constructed, and the full-length VP1 sequence was determined. The phylogenetic tree was constructed by MEGA5 software for phylogenetic analysis. The conserved amino acid sequences of conserved epitopes of VP1 were analyzed by DNAstar software.
