MiR-330-5p和miR-433-3p通過靶向BCAA通路相關(guān)基因?qū)d羊前體脂肪細(xì)胞分化調(diào)控的研究

MicroRNAs(miRNAs)在基因表達(dá)的轉(zhuǎn)錄調(diào)控中起著重要作用。miRNAs缺失可影響脂肪組織的形態(tài)、結(jié)構(gòu)和大小,甚至導(dǎo)致其基本功能的損傷。支鏈氨基酸(BCAA)包括亮氨酸、異亮氨酸和纈氨酸。在脂肪組織中,過多的BCAA攝入會(huì)轉(zhuǎn)化成脂肪儲(chǔ)存起來,脂肪組織有效地把BCAA碳骨架轉(zhuǎn)化成新的脂肪酸,作為脂肪分化和脂肪合成的碳來源,所以BCAA的攝入有利于成脂功能。BCAT2是BCAA代謝過程中的第一個(gè)催化酶,催化BCAA分解為支鏈酮酸的可逆反應(yīng)。脂肪分化過程中,BCAT2還參與支鏈氨基酸的代謝。在后續(xù)分解過程中,支鏈酮酸需在限速酶支鏈酮酸脫氫酶復(fù)合體(BCKDH)作用下才能特異性分解,BCKDHA和BCKDHB是BCKDH E1結(jié)構(gòu)的α和β亞基,它們同樣參與BCAA的代謝與脂肪的代謝。BCAT2,BCKDH 和BCKDHB三者的異常表達(dá),都會(huì)引起B(yǎng)CAA代謝的紊亂、代謝產(chǎn)物的大量積累以及三羧酸循環(huán)過程的障礙,在一定程度上影響了機(jī)體能源物質(zhì)的供應(yīng),甚至?xí)斐梢恍┐x疾病。本研究采用綿羊前體脂肪細(xì)胞為試驗(yàn)對(duì)象,首先對(duì)采取的細(xì)胞進(jìn)行驗(yàn)證,通過對(duì)培養(yǎng)和分化過程中的不同時(shí)段觀察細(xì)胞形態(tài)、油紅O染...
