葛芪復方對仔豬大腸桿菌性腹瀉作用效果研究

【摘要】:葛芪復方是中國農(nóng)業(yè)科學院上海獸醫(yī)研究所藥學研究室自主篩選的新型復方,該復方根據(jù)中獸醫(yī)辯證施治理論,具有良好的抗腹瀉作用效果。為了評價該復方的作用效果,進行了臨床前藥效評價試驗,包括復方對小鼠大腸桿菌性腹瀉作用效果研究,仔豬大腸桿菌性腹瀉病理模型研究,復方對仔豬大腸桿菌性腹瀉作用效果研究以及復方及幾味單藥對大腸桿菌體外抑菌效果研究,為葛芪復方新藥研究提供試驗資料。1復方對小鼠大腸桿菌性腹瀉作用效果研究通過預先建立小鼠大腸桿菌性腹瀉病理模型,設計0.3mL/10g藥物組、0.2mL/10g藥物組、0.1mL/10g藥物組、模型對照組和空白對照組,連續(xù)灌服給藥7d,觀察復方對腹瀉小鼠的防治效果。結(jié)果表明,給藥后各劑量組小鼠平均增重、血液學指標、脾和胸腺臟器指數(shù)與空白對照組無顯著差異,與模型對照組差異明顯(P0.05)。十二指腸組織病理學觀察各劑量組沒有發(fā)現(xiàn)明顯病變,模型對照組腸粘膜絨毛腫脹變粗,由此說明該復方能夠有效防治小鼠腹瀉。2仔豬大腸桿菌性腹瀉病理模型研究通過設計5mL/10Kg感染組、2.5mL/10Kg感染組、1mL/10Kg感染組和空白對照組,各感染組仔豬通過口腔灌服相應的3×109CFU/mL大腸桿菌菌懸液,連續(xù)3d。結(jié)果表明,高劑量組、中劑量組仔豬平均增重、細菌學檢查、血液學指標及電解質(zhì)指標與空白對照組差異顯著(P0.05)。由此說明,通過口腔灌服3×109CFU/mL大腸桿菌菌懸液方式,按照5mL/10Kg劑量,連續(xù)3d,可以有效建立仔豬大腸桿菌性腹瀉模型。3復方對仔豬大腸桿菌性腹瀉作用效果研究通過預先建立仔豬大腸桿菌性腹瀉病理模型,設計10mL/10Kg藥物組、5mL/10Kg藥物組、2.5mL/10Kg藥物組、模型對照組和空白對照組,連續(xù)灌服給藥7d,觀察復方對腹瀉仔豬的防治效果。結(jié)果表明,各劑量組仔豬臨床癥狀、白細胞計數(shù)(WBC)、白球比(A/G)及鈉(Na)、鉀(K)等電解質(zhì)指標與空白對照組無顯著差異,與模型對照組差異明顯(P0.05)。十二指腸組織病理學觀察各劑量組沒有發(fā)現(xiàn)明顯病變,模型對照組腸粘膜上皮及腺體壞死脫落,固有層間質(zhì)中度炎癥細胞增多浸潤,損傷結(jié)構(gòu)沒有恢復,由此說明該復方能有效防治仔豬腹瀉。4復方及幾味單藥對大腸桿菌體外抑菌效果研究試驗采用瓊脂擴散法和微量二倍稀釋法應用復方及幾味單藥水提取物對豬大腸桿菌進行體外抑菌試驗,測定抑菌圈直徑和最小抑菌濃度(MIC)。結(jié)果表明:黃連、黃芩和復方對大腸桿菌敏感,體外抑菌效果比較好。
[Abstract]:GE Qi compound is a new compound selected independently by the Pharmacy Research Department of Shanghai Veterinary Research Institute of Chinese Academy of Agricultural Sciences. It has a good anti-diarrhea effect according to the dialectical treatment theory of Chinese veterinary medicine. In order to evaluate the effect of the compound, the pre-clinical efficacy evaluation test was carried out, including the effect of the compound on the diarrhea induced by Escherichia coli in mice, and the pathological model of colibacillosis in piglets. Study on the effect of compound on diarrhea caused by Escherichia coli in piglets and the bacteriostatic effect of compound and several single drugs on Escherichia coli in vitro. To provide experimental data for the study of new drugs of GE Qi compound. 1 the effect of compound on Escherichia coli diarrhea in mice was studied. By establishing the pathological model of Escherichia coli diarrhea in mice in advance, the 0.3mL/10g drug group and the 0.2mL/10g drug group were designed. 0.1mL/10g drug group, model control group and blank control group were administered continuously for 7 days to observe the preventive and therapeutic effect of compound on diarrhea mice. The results showed that the average weight gain, hematologic index, spleen and thymus organ index of each dose group were not significantly different from those of the blank control group after administration (P0.05). Histopathological observation of duodenum no obvious pathological changes were found in each dose group. The intestinal villi in the model control group became thicker and thicker than those in the control group. 2 the pathological model of colibacillosis in piglets was studied by designing 5mL/10Kg infection group, 2.5mL/10Kg infection group, 1mL/10Kg infection group and blank control group. The piglets of each infected group were given corresponding 3 脳 109CFU/mL Escherichia coli suspensions through oral cavity for 3 days. The results showed that the average weight gain, bacteriological examination, hematological and electrolyte indexes of piglets in high and medium dose groups were significantly different from those in the control group (P0.05). Therefore, by oral administration of 3 脳 109CFU/mL Escherichia coli suspension, according to the dose of 5mL/10Kg, for 3 days, The effect of compound on diarrhea induced by Escherichia coli in piglets was studied. By establishing the pathological model of Escherichia coli diarrhea in piglets in advance, the 10mL/10Kg drug group and the 5mL/10Kg drug group were designed. 2.5mL/10Kg drug group, model control group and blank control group were administered continuously for 7 days to observe the control effect of compound prescription on diarrhea piglets. The results showed that there were no significant differences in the clinical symptoms, white blood cell count (WBC), white ball ratio (A / G) and sodium, (Na), and potassium (K) between the different dosage groups and the blank control group, but there was significant difference between the two groups (P0.05). Duodenal histopathology observation showed no obvious pathological changes in each dose group. The intestinal mucosal epithelium and glands in the model control group were necrotic and exfoliated, the inflammatory cells in the lamina propria were increased and infiltrated, and the injury structure did not recover. The results show that the compound can effectively prevent and cure piglets diarrhea. (4) the study on the bacteriostatic effect of compound and several single drugs on Escherichia coli in vitro; A study on the bacteriostatic effect of compound and several kinds of single medicine extract on pigs by Agar diffusion method and trace double dilution method Bacteriostasis test of Escherichia coli in vitro, Determination of the diameter and minimum inhibitory concentration of bacteriostasis (MIC). The results showed that Coptis chinensis, Radix Scutellariae and compound were sensitive to Escherichia coli and had better bacteriostatic effect in vitro.
