豬Pit-1基因多態(tài)性及其與生長性狀的關(guān)聯(lián)分析

【摘要】:利用現(xiàn)代分子標(biāo)記技術(shù)尋找影響豬生長性狀的標(biāo)記基因座,為應(yīng)用標(biāo)記輔助選擇培育高生產(chǎn)性能豬的品種或品系提供幫助。本試驗采用PCR-RFLP(RsaⅠ限制性內(nèi)切酶)分子標(biāo)記技術(shù),分析了杜洛克、長白、大白及長大二元雜交4個品種共381頭豬Pit-1基因exon 5至intron 5內(nèi)多態(tài)性,并采用最小二乘法分析了Pit-1基因多態(tài)性對生長性狀影響的遺傳效應(yīng)。經(jīng)檢測,4個品種Pit-1基因RsaⅠ位點均存在多態(tài)性。等位基因A的基因頻率分別為0.831 7、0.830 2、0.676 7、0.752 3,A為優(yōu)勢基因。被測豬群Pit-1基因的基因頻率在被測豬群中均處于Hardy-Weinberg平衡狀態(tài)(p0.05)。最小二乘分析表明:Pit-1基因3種基因型的豬在出生重、斷奶重及70日齡重性狀上均呈現(xiàn)BBABAA趨勢(其中,長白豬只存在AB,AA 2種基因型),B為優(yōu)勢基因。Pit-1基因exon 5至intron 5 RsaⅠ位點多態(tài)性對測試豬群生長性狀有一定影響,但是否是影響生長性狀的一個候選基因之一或是與影響豬生長性狀的QTL連鎖的基因,還需進(jìn)一步研究。
