肌醇類(lèi)冰晶抑制劑和海藻糖對(duì)山羊精子冷凍保存效果的影響

【摘要】:為進(jìn)一步降低冷凍解凍過(guò)程對(duì)山羊精子的理化損傷,本試驗(yàn)系統(tǒng)檢驗(yàn)了肌醇類(lèi)冰晶抑制劑(SIB)和海藻糖對(duì)山羊精子的冷凍保護(hù)效果。采用假陰道法采集6只云南黑山羊精液,離心洗滌去除精漿后和冷凍稀釋液混合,經(jīng)4℃平衡、液氮?dú)庀囝A(yù)凍后直接投入液氮保存。解凍后檢測(cè)精子活力、頂體、質(zhì)膜以及低滲耐受性。同時(shí)采用透射電鏡(TEM)觀察冷凍對(duì)精子超微結(jié)構(gòu)的影響。結(jié)果表明:1,4-環(huán)己二醇(1,4-CHD)和海藻糖處理組精子活力(41.35%±8.14%和43.59%±6.23%)、頂體完整性(62.73%±7.62%和64.29%±7.81%)和低滲耐受性(40.29%±7.41%和39.79%±6.58%)均顯著高于對(duì)照組(35.97%±6.21%、55.46%±8.91%和34.17%±5.68%,P0.05),且海藻糖可以有效抑制冷凍解凍過(guò)程導(dǎo)致的精子凋亡。流式分析結(jié)果表明,1,4-CHD不能降低冷凍對(duì)山羊精子質(zhì)膜完整性和磷脂酰絲氨酸(PS)分布的損傷,但是海藻糖可以顯著降低冷凍解凍過(guò)程中對(duì)山羊精子質(zhì)膜的損傷。電鏡結(jié)果表明,冷凍對(duì)精子質(zhì)膜和頂體等結(jié)構(gòu)損傷嚴(yán)重,解凍后部分精子質(zhì)膜發(fā)生腫脹、脫落或斷裂等現(xiàn)象。而且,頂體外膜存在脫落現(xiàn)象,頂體內(nèi)容物發(fā)生部分甚至完全泄漏。電鏡結(jié)果進(jìn)一步證實(shí)海藻糖對(duì)冷凍精子質(zhì)膜的保護(hù)效果要優(yōu)于1,4-環(huán)己二醇?傊,肌醇類(lèi)SIB和海藻糖可以降低山羊精子的冷凍損傷,但相對(duì)于海藻糖而言,肌醇類(lèi)SIB并不能降低冷凍對(duì)山羊精子質(zhì)膜的損傷。
