不同遺傳背景豬群體黏病毒耐藥蛋白1基因多態(tài)性分析

【摘要】:為探討不同遺傳背景豬黏病毒耐藥蛋白1(myxovirus resistance,Mx1)基因的遺傳多態(tài)性,本試驗(yàn)采用RT-PCR方法擴(kuò)增克隆出豬Mx1基因并進(jìn)行生物信息學(xué)分析,采用高分辨率熔解曲線法(HRM)對(duì)民豬、長(zhǎng)白山野豬與民豬雜交豬(簡(jiǎn)稱野雜豬)和大白豬3個(gè)群體Mx1基因外顯子2區(qū)多態(tài)性進(jìn)行分析。結(jié)果顯示,從民豬及野雜豬中成功克隆出4條Mx1基因mRNA序列。該序列含有1個(gè)1 992bp的完整開(kāi)放閱讀框(ORF),編碼663個(gè)氨基酸。比較發(fā)現(xiàn)4條Mx1基因序列中共含有30個(gè)SNPs位點(diǎn),其中17個(gè)SNPs位點(diǎn)引起了氨基酸序列的變異。功能結(jié)構(gòu)域分析發(fā)現(xiàn),大部分氨基酸突變位點(diǎn)位于Mx1基因功能結(jié)構(gòu)域兩側(cè)。HRM多態(tài)性分析顯示,Mx1基因外顯子2區(qū)DD基因型作為優(yōu)勢(shì)基因型在3個(gè)群體中具有最高的基因型頻率,其中大白豬中DD基因型頻率為100%,野雜豬為62.5%,民豬為56.2%;蚨鄳B(tài)性分析還發(fā)現(xiàn)該區(qū)域存在更多的核苷酸及氨基酸突變位點(diǎn),表明民豬和野雜豬Mx1基因存在更豐富的基因多態(tài)性。
[Abstract]:In order to investigate the genetic polymorphism of porcine myxovirus resistance protein 1 (myxovirus resistance,Mx1) gene from different genetic backgrounds, porcine Mx1 gene was amplified by RT-PCR method and analyzed by bioinformatics. High resolution fusion curve method (HRM) was used to analyze the genetic polymorphism of porcine myxovirus resistance protein 1 (myxovirus resistance,Mx1) gene. The polymorphism of exon 2 of Mx1 gene in three populations of Changbai Mountain wild boar and Min pig (wild hybrid pig) and big white pig was analyzed. The results showed that four mRNA sequences of Mx1 gene were cloned successfully from Min pig and wild crossbred pig. The sequence contains a 1 992bp complete open reading frame (ORF), encoding 663 amino acids. A total of 30 SNPs loci were found in 4 Mx1 gene sequences, 17 of which caused amino acid sequence variation. The results of functional domain analysis showed that most of the amino acid mutation loci were located at the two sides of the Mx1 gene functional domain. The results showed that the DD genotype of exon 2 of Mx1 gene had the highest genotype frequency among the three populations. The frequency of DD genotypes in large white pigs, wild crossbred pigs and Min pigs were 100, 62.5 and 56.2, respectively. It was also found that there were more nucleotide and amino acid mutation sites in this region, which indicated that the Mx1 gene of Minzhou pig and wild crossbred pig had more polymorphism.
