【摘要】:目的:通過(guò)體外誘導(dǎo)細(xì)菌產(chǎn)生耐藥性,分析誘導(dǎo)前后細(xì)菌耐藥基因與毒力的變化,探討耐藥基因與毒力之間的關(guān)系,從而為特定耐藥病原體專用新藥的研發(fā)提供科學(xué)依據(jù),亦為鏈球菌產(chǎn)生耐藥性的風(fēng)險(xiǎn)評(píng)估提供試驗(yàn)基礎(chǔ)。方法:本試驗(yàn)采用微量肉湯稀釋法測(cè)定抗微生物藥對(duì)細(xì)菌的最小抑菌濃度(MIC);采用體外逐漸增加藥物濃度的方法對(duì)標(biāo)準(zhǔn)菌株無(wú)乳鏈球菌CVCC1886和停乳鏈球菌CVCC3701進(jìn)行耐藥性誘導(dǎo);采用PCR擴(kuò)增技術(shù)對(duì)誘導(dǎo)耐藥前后的菌株進(jìn)行耐藥基因和毒力基因的檢測(cè);采用形態(tài)學(xué)觀察法對(duì)誘導(dǎo)前后菌株的生理生化性質(zhì)進(jìn)行對(duì)比;通過(guò)測(cè)定受試菌株對(duì)小鼠的LD50比較誘導(dǎo)前后菌株的毒力變化。結(jié)果:1、經(jīng)體外誘導(dǎo),無(wú)乳鏈球菌CVCC1886和停乳鏈球菌CVCC3701對(duì)7種抗菌藥均達(dá)到了耐藥水平;2、經(jīng)PCR檢測(cè),除青霉素誘導(dǎo)的停乳鏈球菌CVCC3701在誘導(dǎo)后耐藥基因和毒力基因均發(fā)生了增加外,用其他抗菌藥誘導(dǎo)前后菌株的耐藥基因和毒力基因均無(wú)變化;3、經(jīng)形態(tài)學(xué)觀察表明CVCC3701-青霉素的生長(zhǎng)繁殖速度明顯低于停乳鏈球菌CVCC3701的生長(zhǎng)繁殖速度;4、小鼠毒力試驗(yàn)表明誘導(dǎo)前停乳鏈球菌CVCC3701和誘導(dǎo)后CVCC3701-青霉素的LD50值分別為5.45x106CFU/mL、5.82x108CFU/mL,誘導(dǎo)后的毒力較誘導(dǎo)前減弱了1.1×102倍,誘導(dǎo)后CVCC3701-青霉素菌株的毒力明顯低于誘導(dǎo)前的停乳鏈球菌CVCC3701。
[Abstract]:Objective: to investigate the relationship between drug resistance genes and virulence by inducing drug resistance of bacteria in vitro, and to provide scientific basis for the development of new drugs for specific drug resistant pathogens. It also provides experimental basis for risk assessment of streptococcus resistance. Methods: the minimal inhibitory concentration (MIC) of antimicrobial agents to bacteria was determined by broth dilution method, and the drug resistance of standard strains CVCC1886 and CVCC3701 was induced by increasing drug concentration in vitro. PCR amplification technique was used to detect the resistance genes and virulence genes of the strains before and after induction of drug resistance, and morphological observation was used to compare the physiological and biochemical properties of the strains before and after induction. The toxicity of the tested strain to LD50 in mice was compared before and after induction. Results: after in vitro induction, both Streptococcus actinomycis CVCC1886 and Streptococcus punctatus CVCC3701 reached the level of resistance to 7 antimicrobial agents, which were detected by PCR. The resistance genes and virulence genes of Streptococcus deltostans CVCC3701 induced by penicillin were increased, but the resistance genes and virulence genes were not changed before and after induced by other antimicrobial agents. 3. Morphological observation showed that the growth and reproduction rate of CVCC3701-penicillin was obviously lower than that of Streptococcus acuminatum CVCC3701. 4. The virulence test of mice showed that the LD50 values of CVCC3701 and CVCC3701-penicillin were 5.45x106CFU / mL5.82x108CFU / mLrespectively, the virulence of induced streptococci CVCC3701-penicillin was 1.1 脳 102.The virulence of induced strain CVCC3701-penicillin was obviously lower than that of CVCC3701before induction.
