【摘要】：本研究旨在利用MiSeq高通量測(cè)序技術(shù)揭示錦江牛瘤胃細(xì)菌組成及多樣性。選用3頭裝有永久性瘤胃瘺管的錦江牛成年公牛[體重(400±20)kg],采集瘤胃液樣品后提取細(xì)菌DNA,對(duì)瘤胃細(xì)菌的16S rDNA序列V4~V5區(qū)進(jìn)行MiSeq測(cè)序,分析物種的豐度、分布及α-多樣性。結(jié)果表明:3個(gè)樣本共獲得56 763條高質(zhì)量16S rDNA序列,946個(gè)分類(lèi)操作單元(OTU),錦江牛瘤胃細(xì)菌豐富度指數(shù)Chao指數(shù)、Ace指數(shù)分別為836、841,α-多樣性指數(shù)Shannon指數(shù)、Simpson指數(shù)分別為4.96、0.021 5;經(jīng)過(guò)物種注釋,15個(gè)門(mén)、23個(gè)綱、26個(gè)科以及44個(gè)屬在所有樣品中被鑒定。在門(mén)水平,擬桿菌門(mén)(Bacteroidetes)(64.37%)、厚壁菌門(mén)(Firmicutes)(21.20%)和變形菌門(mén)(Proteobacteria)(7.59%)數(shù)量最多;在綱水平,主要包括擬桿菌綱(Bacteroidia)(52.88%)和梭菌綱(Clostridia)(17.03%);在科水平,主要包括普雷沃氏菌科(Prevotellaceae)(36.26%)、瘤胃菌科(Ruminococcaceae)(9.33%)和毛螺菌科(Lachnospiraceae)(4.89%);在屬水平,優(yōu)勢(shì)細(xì)菌主要包括普雷沃氏菌屬(Prevotella)(28.97%)、帕拉普氏菌屬(Paraprevotella)(3.08%)、理研菌屬(Rikenella)(2.28%)、梭菌_Ⅳ(ClostridiumⅣ)(1.77%)、解琥珀酸菌屬(Succiniclastium)(1.62%)和瘤胃球菌屬(Ruminococcus)(1.53%)。結(jié)果證明,錦江牛瘤胃細(xì)菌多樣性相對(duì)較低;錦江牛瘤胃中最優(yōu)勢(shì)菌門(mén)是擬桿菌門(mén),其次是厚壁菌門(mén);普雷沃氏菌屬是錦江牛瘤胃中最優(yōu)勢(shì)細(xì)菌屬。
[Abstract]:The aim of this study was to reveal the composition and diversity of rumen bacteria in Jinjiang cattle by using MiSeq high throughput sequencing technique. Three adult bulls with permanent rumen fistula [body weight: 400 鹵20)kg] were collected from rumen fluid to extract bacterial DNA. The 16s rDNA sequence of rumen bacteria was sequenced in V4 / V5 region. The species abundance, distribution and 偽 -diversity were analyzed. The results showed that a total of 56,763 high quality 16s rDNA sequences were obtained from three samples. The results showed that 946 taxonomic operating units were obtained from the three samples. The rumen bacterial richness index (Chao) and 偽 -diversity index (Shannon index and Simpson index) of Jinjiang cattle were 836 841 and 4.96 0. 021 5, respectively.
