【摘要】：以病理組織學(xué)觀察為基礎(chǔ),建立檢測綿羊肺炎支原體的間接免疫組織化學(xué)和間接免疫熒光方法,以綿羊肺炎支原體標(biāo)準(zhǔn)血清為一抗,分別以家兔抗山羊Ig G-HRP和家兔抗山羊Ig G-FITC為二抗,對綿羊肺炎支原體陽性肺組織切片及陰性對照組切片進(jìn)行HE染色、免疫組織化學(xué)染色及間接免疫熒光染色,并優(yōu)化染色條件,以確保組織切片在最佳的條件下進(jìn)行鑒定。結(jié)果顯示,本試驗(yàn)所建立的免疫組織化學(xué)方法與PCR方法對病樣的檢出率相符,且高于傳統(tǒng)的分離培養(yǎng)法。結(jié)果表明,本試驗(yàn)建立的間接免疫組織化學(xué)方法和間接免疫熒光方法可用于對臨床病例的組織樣本檢測,具有很強(qiáng)的特異性和可靠性,可直觀地觀察和探究該病原在機(jī)體內(nèi)的定位、動態(tài)分布以及致病機(jī)理,以期為臨床診斷提供更可靠的方法。
[Abstract]:On the basis of histopathological observation, an indirect immunohistochemical and indirect immunofluorescence method for the detection of Mycoplasma pneumoniae in sheep was established. The standard serum of Mycoplasma pneumoniae was used as the first antibody. Using rabbit anti-goat Ig G-HRP and rabbit anti-goat Ig G-FITC as second antibodies, HE staining, immunohistochemical staining and indirect immunofluorescence staining were performed on the positive lung tissue sections of sheep mycoplasma pneumoniae and negative control groups, and the staining conditions were optimized. To ensure that tissue sections are identified under optimal conditions. The results showed that the immunocytochemical method established in this study was consistent with the detection rate of PCR method and was higher than that of the traditional isolation culture method. The results show that the indirect immunohistochemical method and indirect immunofluorescence method established in this study can be used to detect the tissue samples of clinical cases, and have strong specificity and reliability. The location, dynamic distribution and pathogenesis of the pathogen in the organism can be observed and explored intuitively in order to provide a more reliable method for clinical diagnosis.
