【摘要】：為建立準(zhǔn)確、快速檢測(cè)兔支氣管敗血波氏桿菌(Bordetella bronchiseptica)的PCR方法,根據(jù)Gen Bank中波氏桿菌ptxA基因設(shè)計(jì)1對(duì)特異性引物,用建立的PCR方法擴(kuò)增兔波氏桿菌,可擴(kuò)增出870 bp的目的片段。該方法特異性好,僅對(duì)波氏桿菌有特異性的擴(kuò)增,而對(duì)大腸埃希菌、無(wú)乳鏈球菌、金黃色葡萄球菌、多殺性巴氏桿菌等病原菌的擴(kuò)增結(jié)果呈陰性;敏感性較高,最低檢測(cè)限可達(dá)6.8×10~(-7)μg/m L;用建立的PCR方法檢測(cè)臨床疑似感染支氣管敗血波氏桿菌病兔的臟器及鼻拭子樣品,可擴(kuò)增出目的條帶,且與細(xì)菌分離的結(jié)果一致�？梢�(jiàn),建立的檢測(cè)方法敏感、特異,可用于臨床上兔波氏桿菌病的快速診斷及鑒定。
[Abstract]:In order to establish a PCR method for rapid detection of Pasteurella bronchiolus (Bordetella bronchiseptica), 1 pairs of specific primers were designed according to the ptxA gene of Pasteurella in Gen Bank. The amplified fragment of Pasteurella rabbit was amplified by the established PCR method, and the target fragment of 870 BP was amplified. This method has a good specificity and only specific expansion of Pasteurella. The amplification results of Escherichia coli, Streptococcus free Streptococcus, Staphylococcus aureus and Pasteurella multocida were negative, and the sensitivity was higher, the minimum detection limit was 6.8 * 10~ (-7) g/m L, and the PCR method was used to detect the viscera and nose swab samples of clinically suspected bronchobacillosis rabbits. The results showed that the established method is sensitive and specific, and can be used for the rapid diagnosis and identification of Bordetella in rabbits.
