【摘要】：【目的】Dmrt7基因是Dmrt基因家族成員之一,影響公畜精子形成。迄今,有關(guān)水牛精子發(fā)生的遺傳基礎(chǔ)的研究很少,本研究旨在對(duì)檳榔江水牛Dmrt7基因完整CDS區(qū)進(jìn)行克隆和組織差異表達(dá)分析�！痉椒ā坎捎肦T-PCR和功能生物信息學(xué)方法。【結(jié)果】檳榔江水牛Dmrt7基因完整編碼區(qū)序列長(zhǎng)1 113 bp,編碼370個(gè)氨基酸;Dmrt7包含DM和DMRT-like兩個(gè)保守結(jié)構(gòu)域,無(wú)信號(hào)肽,無(wú)跨膜結(jié)構(gòu),為核內(nèi)蛋白。同源性分析顯示,水牛與其他牛科物種Dmrt7氨基酸序列相似度達(dá)95%以上。在檢測(cè)的10個(gè)組織中,Dmrt7基因僅在水牛睪丸組織中表達(dá)且高表達(dá)。在第6外顯子發(fā)現(xiàn)1個(gè)SNP位點(diǎn)(c.665AG),引起了p.Q222R的替換,此位點(diǎn)的替換對(duì)Dmrt7蛋白功能無(wú)影響。【結(jié)論】有助于了解水牛Dmrt7基因的基本功能。
[Abstract]:[objective] Dmrt7 gene is a member of Dmrt gene family, which affects sperm formation in male animals.To date, little research has been done on the genetic basis of spermatogenesis in buffalo.The purpose of this study was to clone the complete CDS region of the Dmrt7 gene and analyze the tissue differential expression of the complete CDS region of the Dmrt7 gene of the water buffalo. [methods] RT-PCR and functional bioinformatics methods were used. [results] the complete coding region of the Dmrt7 gene of the water buffalo was long.1,113bp, encoding 370 amino acids Dmrt7 containing two conserved domains DM and DMRT-like.No signal peptide, no transmembrane structure, is a nuclear protein.Homology analysis showed that the similarity of Dmrt7 amino acid sequence between buffalo and other bovine species was more than 95%.The Dmrt7 gene was only expressed and overexpressed in the testis of buffalo.A SNP locus named c. 665AGN was found at exon 6, which led to the replacement of p.Q222R, which had no effect on the function of Dmrt7 protein. [conclusion] it is helpful to understand the basic function of buffalo Dmrt7 gene.
