東北虎源蛔蟲(chóng)線粒體cox1基因序列進(jìn)化分析研究

【摘要】:為了解東北虎源蛔蟲(chóng)與其他蛔蟲(chóng)的種群遺傳關(guān)系,本研究對(duì)湖南省長(zhǎng)沙生態(tài)動(dòng)物園東北虎糞便中采集的蛔蟲(chóng)成蟲(chóng)的線粒體DNA中細(xì)胞色素I(cox1)基因序列進(jìn)行擴(kuò)增,將測(cè)序結(jié)果與GenB ank中登錄的其他蛔蟲(chóng)cox1序列進(jìn)行對(duì)比及種系發(fā)育樹(shù)分析。結(jié)果顯示,來(lái)自長(zhǎng)沙生態(tài)動(dòng)物園7個(gè)東北虎蛔蟲(chóng)樣品cox1序列長(zhǎng)度基本一致,為413 bp~414 bp,與貓弓首蛔蟲(chóng)同源性最高,達(dá)98.4%以上,而且系統(tǒng)進(jìn)化分析顯示所有樣品均與貓弓首蛔蟲(chóng)位于同一分支。該結(jié)果表明,pcox1序列種內(nèi)相對(duì)保守,種間差異較大,因此可以作為貓科弓首蛔蟲(chóng)的種間遺傳變異研究的分子遺傳標(biāo)記,從而為蛔蟲(chóng)的群體遺傳研究奠定了基礎(chǔ)。
[Abstract]:In order to understand the genetic relationship between Ascaris ricoides and other Ascaris, we amplified the cytochrome Icox1 gene from mitochondrial DNA of adult Ascaris lumbricoides collected from the feces of Amur tiger in Changsha Ecological Zoo of Hunan Province.The sequencing results were compared with the cox1 sequences of other Ascaris lumbricoides registered in GenB ank and the phylogenetic tree analysis.The results showed that the length of cox1 sequence of 7 Amur tiger ascariasis samples from Changsha Ecological Zoo was basically the same, which was 413 bp~414 bp. the homology was the highest with Toxocariasis catnii, which was above 98.4%.Phylogenetic analysis showed that all samples were located in the same branch as Ascaris catacaroides.The results showed that the pcox1 sequence was relatively conserved within species and varied greatly among species, so it could be used as a molecular genetic marker for the study of genetic variation of Ascaris lumbricoides among species, thus laying a foundation for the population genetic study of Ascaris lumbricoides.
