仔豬均勻度遺傳參數(shù)估計(jì)及仔豬初生重與血清皮質(zhì)醇、胰島素含量的相關(guān)性研究

【摘要】:研究顯示,仔豬均勻度對養(yǎng)豬業(yè)有極大影響,血清皮質(zhì)醇及胰島素含量可能與仔豬的初生重相關(guān)。本研究采用動(dòng)物模型REML(約束最大似然)方法對大白豬8個(gè)繁殖性狀的遺傳參數(shù)進(jìn)行估計(jì),利用SAS9.1統(tǒng)計(jì)軟件的GLM程序分析年份、配種季節(jié)、胎次對8個(gè)繁殖性狀的影響,采用Duncan,s法進(jìn)行各固定效應(yīng)間繁殖性狀的多重比較;采用放射免疫分析法檢測不同初生重仔豬血清皮質(zhì)醇含量及胰島素含量。旨在為育種工作提供仔豬均勻度遺傳參數(shù)數(shù)據(jù)支持;為探索皮質(zhì)醇及胰島素對初生重的影響起參考作用。1.仔豬均勻度遺傳參數(shù)估計(jì)利用5種統(tǒng)計(jì)模型對大白豬窩內(nèi)仔豬初生重變異系數(shù)(CV0)、窩內(nèi)仔豬21日齡重變異系數(shù)(CV3)、總產(chǎn)仔數(shù)(TNB)、產(chǎn)活仔數(shù)(NBA)、產(chǎn)死仔數(shù)(NBD)、初生窩重(TLW0)、21日齡窩重(TLW3)及妊娠周期(GL)進(jìn)行遺傳參數(shù)估算。經(jīng)比較,應(yīng)用模型V(包括個(gè)體加性遺傳效應(yīng),母體遺傳效應(yīng)、永久環(huán)境效應(yīng)、與配公豬效應(yīng)、母豬所在窩效應(yīng))得到的殘差方差所占比例小。因此本實(shí)驗(yàn)采用模型V進(jìn)行繁殖性狀遺傳參數(shù)估計(jì)。遺傳參數(shù)估計(jì)結(jié)果表明:TNB,NBA,NBD,CV0,CV3,TLW0,TLW3,GL遺傳力分別為0.14,0.13,0.07,0.07,0.02,0.11,0.08,0.09。TNB與NBA、TNB與TLW0、TNB與TLW3、NBA與TLW0、NBA與TLW3、CV0與CV3、TLW0與TLW3之間存在明顯的遺傳正相關(guān)(遺傳相關(guān)分別為0.93,0.61,0.61,0.65,0.73,0.65,0.91);TNB與CV0、TNB與CV3、CV0與NBD之間遺傳相關(guān)分別為0.29,0.17,0.22。固定效應(yīng)方差分析表明:年份對TNB和CV0影響不顯著(P0.05),對其余性狀的影響都達(dá)到極顯著(P0.01);配種季節(jié)對TNB、NBA、CV3影響不顯著(P0.05),對CV0影響達(dá)顯著水平(P0.05),對TLW0,TLW3和GL影響達(dá)到極顯著水平(P0.01);胎次對對TNB、NBA、NBD的影響達(dá)顯著水平(P0.05),對CV0、CV3、TLW0、TLW3及GL影響極顯著(P0.01)。產(chǎn)仔數(shù)對所有性狀影響都達(dá)到極顯著水平(P0.01)。繁殖性狀最小二乘均值表明:隨著胎次增加,TNB、NBA、TLW0、TLW3呈現(xiàn)出相同的變化規(guī)律,第一胎最低,以后逐漸上升,到3-4胎時(shí)候達(dá)到最大,然后又開始下降。季節(jié)對繁殖性狀的影響表現(xiàn)為冬季生產(chǎn)性能最佳;隨著產(chǎn)仔數(shù)的增加,CV0、CV3、NBD也增加,而MBW0、MBW3、GL卻下降;隨著產(chǎn)仔數(shù)增加,初生重小于1 Kg仔豬所占比例增加,體重大于1.8 Kg仔豬所占比例下降。2.血清皮質(zhì)醇和胰島素含量水平與仔豬初生重相關(guān)性分析實(shí)驗(yàn)選取10窩大白豬,每窩包括2頭低初生重(1.0 Kg)2頭高初生重(1.2 Kg),共40頭初生仔豬作為實(shí)驗(yàn)樣本。仔豬出生后前腔靜脈采血,離心后提取血清,采用放射免疫分析法檢測血清中皮質(zhì)醇含量及胰島素含量。研究結(jié)果表明:高出生重組和低出生重組血清皮質(zhì)醇量分別為214.72(ng/ml),267.87(ng/ml),兩組之間差異極顯著(P=0.0083);高出生重組和低出生重組血清胰島素含量分別為28.67(m IU/L),20.92(m IU/L),兩組間差異顯著(P=0.0284)。血清皮質(zhì)醇含量與初生體重之間存在顯著負(fù)相關(guān)(rg=-0.32),血清胰島素含量與體重之間存在正相關(guān)(rg=0.23),但是二者之間相關(guān)性不顯著(P=0.09)。實(shí)驗(yàn)結(jié)果為探索皮質(zhì)醇及胰島素對初生重的影響提供參考。
[Abstract]:Research shows that piglet uniformity to the pig industry has great influence, and the birth weight of piglets serum cortisol and insulin content. This study used the animal model of REML (restricted maximum likelihood) genetic parameters of 8 reproductive traits in large white pigs estimation method, analysis of the year, the use of SAS9.1 statistical software GLM program of breeding season. Effect of parity on 8 reproductive traits by using Duncan, s multiple comparison method of reproductive traits of fixed effects; radioimmunoassay was used to measure the different birth weight of piglets serum cortisol and insulin content. In order to provide piglets for breeding data uniformity and genetic parameters of support; to explore the effects of insulin and cortisol the birth weight of the piglets reference.1. uniformity of genetic parameter estimation of large white nest piglets weight variation coefficient using 5 statistical model (CV0), wo Zi 21 day old pig weight coefficient of variation (CV3), total litter size (TNB), number born alive (NBA), produce litter (NBD), birth weight (TLW0), litter weight at 21 days (TLW3) and pregnancy period (GL) of genetic parameter estimation. By comparison, application the V model (including individual additive genetic effects, maternal genetic effect and permanent environmental effect, and with the boar sows effect, where litter effect) residual variance obtained small proportion. Therefore this experiment adopts the V model to reproduce the genetic parameters estimation of genetic parameters of traits. The results showed that: TNB, NBA, NBD, CV0. CV3, TLW0, TLW3, GL and 0.14,0.13,0.07,0.07,0.02,0.11,0.08,0.09.TNB respectively, heritability of NBA, TNB and TLW0, TNB and TLW3, NBA and TLW0, NBA and TLW3, CV0 and CV3, there were significant genetic positive correlation between TLW0 and TLW3 (genetic correlation = 0.93,0.61,0.61,0.65,0.73,0.65,0.91); TNB and CV0, TNB and CV3, between CV0 and NBD genetic correlation respectively. Show that the 0.29,0.17,0.22. variance analysis of fixed effects: the effect of TNB and CV0 years (P0.05), no significant effects on other traits were at significant (P0.01); breeding season on TNB, NBA, CV3 had no significant effects on CV0 (P0.05), significant effects (P0.05), TLW0, TLW3 and GL influence reached a significant level (P0.01); parity on to TNB, NBA, NBD significant effects (P0.05), CV0, CV3, TLW0, TLW3 and GL had significant effect (P0.01). Litter size were all highly significant for all traits affecting reproductive traits (P0.01) showed that the least squares mean. With the increase of TNB, NBA, parity, TLW0, TLW3 showed the same variation, the first minimum, after rising to 3-4 when the tire reaches the maximum, and then began to decline. The seasonal effect on reproductive traits for winter production best; with the increase of litter size of CV0, CV3, NBD increased, while MBW0, M BW3, GL decreased; with the increase of litter size, birth weight less than 1 Kg increase in the proportion of piglets, piglets weight greater than 1.8 Kg the proportion of.2. analysis of serum cortisol and insulin levels and birth weight of the piglets correlation experiment selected 10 large white nest down, including 2 low birth weight per litter (1 Kg) 2 heads the birth weight (1.2 Kg), a total of 40 newborn piglets as the experimental samples. The piglets born after blood samples were collected after centrifugation, extract serum, cortisol and insulin content in serum was measured by radioimmunoassay. The analysis results show that the high born restructuring and low birth weight were 214.72 recombinant serum cortisol (ng/ml) (ng/ml, 267.87), significant differences between the two groups (P=0.0083); high birth and low birth of recombinant insulin in recombinant were 28.67 (m IU/L), 20.92 (m, IU/L) between the two groups were significant difference (P=0.0284). The content of serum cortisol There was a significant negative correlation between birth weight and body weight (rg=-0.32). There was a positive correlation between serum insulin and body weight (rg=0.23), but the correlation between the two groups was not significant (P=0.09). The results of the experiment provide a reference for exploring the influence of cortisol and insulin on the birth weight.
