【摘要】：為了快速檢測(cè)臨床上副豬嗜血桿菌(Haemophilus parasuis,Hps)感染,根據(jù)Gen Bank中Hps的16S rRNA基因保守區(qū)序列,設(shè)計(jì)1對(duì)特異性引物,利用PCR方法檢測(cè)Hps,并進(jìn)行特異性、敏感性及臨床檢測(cè)。結(jié)果顯示:PCR反應(yīng)選擇58℃為最適退火溫度,0.5μL(20μmol/L)為最適引物量,模板濃度為7.2×10~(-6)μg/mL時(shí)即可擴(kuò)增出814 bp特異性目的片段,與細(xì)菌分離結(jié)果一致。結(jié)果表明:該方法敏感、快速、特異,可用于臨床上Hps感染的快速檢測(cè)。
