【摘要】：新城疫病毒樣顆粒(NDV VLPs)是由基質(zhì)蛋白(M)為病毒骨架,裝配血凝素-神經(jīng)氨酸酶和/或融合蛋白等。已有研究表明,NDV VLPs能誘導(dǎo)機(jī)體產(chǎn)生特異性體液和細(xì)胞免疫應(yīng)答。然而,關(guān)于NDV VLPs如何激活天然免疫應(yīng)答的研究尚無(wú)報(bào)道。樹(shù)突狀細(xì)胞(DCs)作為專職抗原遞呈細(xì)胞,在連接天然免疫與適應(yīng)性免疫之間具有獨(dú)特功能。本試驗(yàn)以NDV VLPs與小鼠DCs為研究靶點(diǎn),將NDV VLPs(M+HN)刺激DC以評(píng)價(jià)DC成熟表征。結(jié)果顯示該顆粒可被DC有效吞噬并遞呈給初始型T細(xì)胞,誘導(dǎo)DC表面MHCⅡ和共刺激分子顯著上調(diào)以及促進(jìn)DC分泌促炎性細(xì)胞因子。另外,還發(fā)現(xiàn)不同組裝類型的NDV VLPs對(duì)誘導(dǎo)DC分泌炎性細(xì)胞因子水平不一。結(jié)果表明,NDV VLPs可誘導(dǎo)DC成熟,這對(duì)更好地理解VLPs激活的天然免疫應(yīng)答奠定基礎(chǔ),并且為優(yōu)選NDV VLPs候選株提供參考依據(jù)。
[Abstract]:NDV-like particles of Newcastle disease virus (NDVVLPs) are composed of matrix protein (M) as viral skeleton, which assembles hemagglutinin-neuraminidase and / or fusion proteins. It has been shown that NDV-like VLPs can induce specific humoral and cellular immune responses. There are no reports on how NDV VLPs activates innate immune response. Dendritic cells (DC), as specialized antigen presenting cells, have unique function in linking innate and adaptive immunity. NDV VLPs and mouse DCs were used as targets in this study. NDV VLPs(M HN) was used to stimulate DC to evaluate DC maturation characterization. The results showed that the granules could be effectively phagocytized by DC and presented to initial T cells, induce MHC 鈪，
