【摘要】：豬流行性腹瀉病毒(PEDV)是引起豬(特別是新生仔豬)急性、高度傳染性消化道疾病的主要病原之一;2010年底以來,豬流行性腹瀉(PED)的再次暴發(fā)給我國乃至全球養(yǎng)豬業(yè)造成了巨大經(jīng)濟損失。最近,研究者已先后建立了基于靶向RNA重組技術(shù)、細菌人工染色體(BAC)載體系統(tǒng)和體外連接技術(shù)的PEDV反向遺傳學操作技術(shù),為PEDV編碼的蛋白質(zhì)的功能、致病機制以及新型疫苗的研發(fā)開辟了新的思路。本文對PEDV反向遺傳操作技術(shù)的研究進展及其在PEDV胰酶依賴性、S蛋白和ORF3蛋白的功能以及新一代疫苗研制等方面的應用現(xiàn)狀進行了綜述與展望。
[Abstract]:Porcine epidemic diarrhea virus (PEDVV) is one of the main pathogens causing acute and highly infectious digestive tract diseases in pigs (especially newborn piglets) since the end of 2010. The repeated outbreak of porcine epidemic diarrhea (PED) has caused huge economic losses to the pig industry in China and the world. Recently, researchers have established a recombinant technology based on targeted RNA. The PEDV reverse genetic manipulation of bacterial artificial chromosome PEDV vector system and in vitro ligation technique is the function of protein encoded by PEDV. The mechanism of pathogenesis and the research and development of new vaccines have opened up a new way of thinking. In this paper, the research progress of PEDV reverse genetic manipulation and its function in PEDV trypsin dependent protein S and ORF3 protein, as well as the development of new generation vaccine, etc. The present situation of application is summarized and prospected.
