肉雞腹脂雙向選擇系中RNA編輯位點的鑒定研究

【摘要】:旨在準確獲得肉雞腹脂雙向選擇系脂肪組織中的RNA編輯位點信息,進而為脂肪相關研究提供更多的信息來源。本研究以東北農業(yè)大學肉雞高、低腹脂雙向選擇品系第十九世代肉雞為試驗材料,使用轉錄組測序(RNA-seq)和基因組測序數據識別肉雞腹部脂肪組織RNA編輯位點。通過生物信息學手段,設定嚴格的數據過濾與篩選策略,構建RNA編輯位點候選集。結果表明,在候選集中,通過對RNA編輯位點的分類和注釋,在低脂系肉雞脂肪組織中發(fā)掘轉換型RNA編輯位點28個,顛換型RNA編輯位點101個,插入缺失型RNA編輯位點71個;在高脂系肉雞脂肪組織中,發(fā)掘轉換型RNA編輯位點30個,顛換型RNA編輯位點84個,插入缺失型RNA編輯位點77個。絕大多數RNA編輯位點落在內含子、基因間區(qū)域及基因上游區(qū)域(內含子區(qū)41.91%,基因間區(qū)域33.08%,基因下游20.59%,基因上游2.94%和外顯子區(qū)1.47%)。試驗驗證了4個候選RNA編輯位點,其中3個位點落在與脂肪形成有重要關系的ATP1B3、SLC6A6和FLNB基因區(qū)域。本研究鑒定和驗證了存在于肉雞脂肪組織中的RNA編輯位點,為進一步研究RNA編輯影響脂肪組織生長發(fā)育的分子機制奠定了基礎。
[Abstract]:The aim of this study was to obtain accurate information of RNA editing site in adipose tissue of bidirectional selection line of abdominal fat in broilers, and to provide more information sources for adipose related studies. Low abdominal fat bidirectional selection of the 19th generation broilers as experimental materials, using RNA-seq) and genomic sequencing data to identify RNA editing sites in abdominal adipose tissue of broilers. Strict data filtering and screening strategies were set up to construct the candidate set of RNA editing sites. The results showed that, by classifying and annotating RNA editing sites, 28 transformative RNA editing sites were found in adipose tissue of low-fat broilers. Among the adipose tissues of high-fat broilers, 30 were found to be transversive RNA editing sites, 84 were transversive RNA editing sites, and 101 were transversive RNA editing sites and 71 insertion deletion RNA editing sites, respectively. The majority of RNA editing sites fell in introns. Intergenic region and gene upstream region (intron region 41.91, intergenic region 33.08, downstream gene 20.5959, upstream 2.94% and exon 1.4775). Four candidate RNA editing sites were tested. Three of the loci were located in the region of ATP-1B3OSLC6A6 and FLNB gene, which were related to adipogenesis. The RNA editing site in adipose tissue of broiler was identified and verified. It provides a basis for further study on the molecular mechanism of RNA editing affecting the growth and development of adipose tissue.
