和田羊與策勒黑羊血液中Mel變化規(guī)律和卵巢子宮MTRN1A的表達(dá)研究

【摘要】:新疆和田地區(qū)的策勒黑羊與和田羊是當(dāng)?shù)刂饕蚍N,前者具有性成熟早、全年發(fā)情和密集產(chǎn)羔的特點(diǎn),后者為半粗毛羊,羊毛品質(zhì)好,但繁殖率低,為季節(jié)性發(fā)情動(dòng)物。有研究報(bào)道,褪黑激素(melatonin,Mel)在一定程度上能對(duì)動(dòng)物繁殖性狀產(chǎn)生影響,本試驗(yàn)旨在通過(guò)對(duì)不同品種綿羊褪黑激素與褪黑激素受體表達(dá)的時(shí)空互作關(guān)系的研究,探討Mel對(duì)和田羊與策勒黑羊繁育機(jī)能的影響。本次試驗(yàn)采集年周期下春分日、夏至日、秋分日和冬至日四個(gè)節(jié)氣當(dāng)日早晨、中午、傍晚和凌晨四個(gè)時(shí)間點(diǎn)的血液,并通過(guò)ELISA法測(cè)定每個(gè)時(shí)間點(diǎn)血液中Mel濃度,同時(shí)采集和田羊與策勒黑羊的卵巢和子宮組織并提取RNA,采用半定量RT-PCR技術(shù)和免疫組化方法,對(duì)年周期下和田羊與策勒黑羊Mel分泌、褪黑激素受體(MTRN1A)基因在mRNA水平與蛋白水平的表達(dá)情況進(jìn)行研究。結(jié)果表明:日周期下,策勒黑羊血液中Mel濃度在春分當(dāng)日中午最低,顯著低于早晨、傍晚和凌晨(P0.05),傍晚最高,但差異不顯著(P0.05);和田羊最高值與最低值出現(xiàn)時(shí)間同策勒黑羊,但四個(gè)時(shí)間點(diǎn)兩兩之間的差異并不顯著(P0.05)。夏至日兩種綿羊均是傍晚最高、早晨最低,但差異并不顯著(P0.05);秋分日策勒黑羊凌晨濃度顯著高于其余時(shí)間(P0.05),早晨、中午差異不顯著(P0.05),但二者顯著低于傍晚(P0.05);和田羊總體變化與策勒黑羊相同,但四個(gè)時(shí)間點(diǎn)之間并無(wú)顯著差異(P0.05)。冬至日兩種綿羊傍晚含量顯著高于其它時(shí)間點(diǎn)(P0.05),早晨、凌晨之間差異不顯著(P0.05),但均顯著高于中午(P0.05)。年周期下,策勒黑羊春分Mel濃度顯著高于夏至與秋分(P0.05),秋分顯著低于冬至(P0.05),夏至顯著低于秋分、冬至(P0.05);同時(shí),春分日卵巢中MTRN1A在mRNA和蛋白水平上,表達(dá)量均最高,夏至則最低,顯著低于冬至和秋分(P0.05),同時(shí)顯著低于春分(P0.05),受體表達(dá)量則是秋分最低,顯著低于冬至(P0.05),但與夏至日差異不顯著(P0.05)。子宮中MTRN1A在基因與蛋白均在夏至日表達(dá)量最低,且與秋分日并無(wú)顯著差異(P0.05),但顯著低于春分(P0.05),三個(gè)季節(jié)同時(shí)顯著低于冬至(P0.05)蛋白表達(dá)量。而和田羊則是血液Mel含量冬至顯著高于夏至與秋分(P0.05),與春風(fēng)差異并不顯著(P0.05),夏至最低,與秋分差異并不顯著(P0.05),但二者顯著低于春分(P0.05);卵巢中MTRN1A基因表達(dá)量在冬至最高,顯著高于春分與夏至(P0.05),與秋分差異并不顯著(P0.05),夏至日最低,顯著低于春分(P0.05)MTRN1A蛋白的表達(dá)量夏至最低,與秋分差異不顯著(P0.05),但二者顯著低于春分(P0.05),同時(shí),三者顯著低于冬至(P0.05);和田羊子宮內(nèi)MTRN1A的基因與受體表達(dá)規(guī)律均與策勒黑羊相同。研究表明,兩種綿羊血液中Mel含量均存在明顯的晝夜節(jié)律和季節(jié)性節(jié)律,但策勒黑羊血液中Mel總體含量高于和田羊,且晝夜變化較大;同樣,兩種綿羊卵巢和子宮組織中MTRN1A基因與受體的表達(dá)均呈現(xiàn)先降后升的季節(jié)性變化規(guī)律,春分日策勒黑羊卵巢中基因表達(dá)量顯著高于和田羊,秋分則相反,其他季節(jié)無(wú)顯著差異;同時(shí)子宮中的mRNA表達(dá)也無(wú)顯著不同。同樣,策勒黑羊春分日卵巢中MTRN1A受體的表達(dá)顯著高于和田羊,冬至日則相反,其余季節(jié)并無(wú)顯著差異;春分日和田羊子宮中受體的表達(dá)顯著高于策勒黑羊,其余季節(jié)則無(wú)顯著差異。說(shuō)明年周期下血液中Mel濃度、MTRN1A基因與蛋白表達(dá)量的節(jié)律性變化規(guī)律,因品種、組織不同而異。本次試驗(yàn)通過(guò)對(duì)策勒黑羊與和田羊血液中Mel含量變化、子宮卵巢中MTRN1A在m RNA水平和蛋白水平表達(dá)的動(dòng)態(tài)規(guī)律研究,旨在為研究Mel對(duì)不同品種綿羊繁育機(jī)能的影響提供一定參考,對(duì)實(shí)際生產(chǎn)中,規(guī);B(yǎng)殖場(chǎng)的飼養(yǎng)管理提供一定的科學(xué)依據(jù)。
[Abstract]:Xinjiang Hetian area of Qira black sheep and Hetian Sheep are the main local sheep, the former has early sexual maturity, characteristics of annual estrus and intensive lambing, the latter half coarse wool sheep, wool quality is good, but the low reproductive rate, seasonal oestrus. Animal studies have reported that melatonin (melatonin, Mel) can have an impact on animal reproductive traits to a certain extent, this experiment aims to study the relationship between the interaction of melatonin and melatonin receptor expression in temporal sheep of different varieties, the effect of Mel on the Hotan sheep and Qira black goat breeding function. The influence of the test collection cycle years the spring equinox, summer solstice, four solar term the autumnal equinox and winter solstice day morning, noon, evening and at four time points of the blood, and through the ELISA method for the determination of Mel concentration in each time point in the blood, while collecting the Hetian Sheep and Qira black goat ovaries and uterus tissue and extract RNA, By using semi quantitative RT-PCR technique and immunohistochemical method, the annual cycle under the Hotan sheep and Qira black sheep Mel secretion of melatonin receptor (MTRN1A) gene expression in mRNA level and protein level. The results showed that the diurnal cycle of Qira black sheep, in the blood concentration of Mel in the first day of spring was lowest at noon, below the morning, evening and morning (P0.05), the highest in the evening, but the difference was not significant (P0.05); and the highest value of the same time Hetian Sheep of Qira black sheep and the lowest value, but no significant difference between the four time points of 22 (P0.05). The study shows that the Mel contents of two sheep blood existed in the circadian rhythm and seasonal rhythm is obvious, but the Qira black sheep blood total Mel was higher than that in Hetian Sheep, and day and night Large changes; similarly, the expression of MTRN1A two gene in sheep ovarian and uterine tissue and receptor showed seasonal variation rose after the first drop, the gene expression of the equinox of Qira black sheep ovarian volume was significantly higher than that of Hetian Sheep, the contrary, no significant difference in other seasons; at the same time, the expression of mRNA in uterus no significant different. Similarly, the expression of MTRN1A receptor of Qira black sheep ovary was significantly higher than that of the spring equinox and winter solstice Tian sheep, on the contrary, there is no significant difference in other seasons; spring equinox and receptor expression in the uterus of Hetian Sheep was significantly higher than that of Qira black sheep, there was no significant difference in other seasons. Through this experiment of Qira black sheep and Hetian Sheep Blood Mel content change, ovary and uterus in M RNA and MTRN1A The study of the dynamic regulation of protein level aims to provide some references for the study of the effects of Mel on the breeding performance of different breeds of sheep, and provide a scientific basis for the breeding and management of large scale farms in actual production.
