血吸蟲果糖二磷酸醛縮酶誘導Th2免疫偏移的研究

【摘要】:目的分析日本血吸蟲感染以及蟲源性糖蛋白果糖二磷酸醛縮酶(Sj FBPA)刺激后小鼠脾臟細胞中白介素4(IL-4)、白細胞分化抗原CD8a因子表達量的動態(tài)變化,并探討Sj FBPA與輔助型T細胞(Th2)免疫應答的關系。方法血吸蟲尾蚴感染C57小鼠10只,設正常對照組。感染8周后處死小鼠,取脾臟制備淋巴細胞懸液;用藻紅蛋白(PE)標記CD8a抗體,藻青蛋白(APC)標記IL-4抗體進行細胞染色;流式細胞儀檢測細胞因子表達量的變化。10只正常C57小鼠處死后取脾臟制成淋巴細胞懸液,加Sj FBPA蛋白體外刺激,按上述流程刺激方法檢測細胞。結果小鼠感染血吸蟲8周后脾細胞中IL-4、CD8a的表達量分別為(22.39±5.93)%、(11.34±2.15)%,正常對照組脾細胞中IL-4、CD8a的表達量分別為(1.39±0.52)%、(1.98±0.84)%,差異有統(tǒng)計學意義(P0.01)。Sj FBPA體外刺激小鼠脾細胞的IL-4、CD8a的表達量分別為(14.14±1.16)%、(8.99±0.81)%,正常對照組脾細胞中IL-4、CD8a的表達量分別為(1.86±0.93)%、(1.69±0.56)%,差異有統(tǒng)計學意義(P0.01)。結論 Sj FBPA在血吸蟲感染中具有誘導Th2免疫偏移的作用,樹突狀細胞作為提呈細胞參與該免疫反應。
[Abstract]:Objective to analyze the dynamic changes of expression of interleukin 4 (IL-4) and leukocyte differentiation antigen (CD8a) in spleen cells of mice after Schistosoma japonicum infection and (Sj FBPA) stimulation with insect-derived glycoprotein fructose diphosphate aldolase (Sj FBPA). To investigate the relationship between Sj FBPA and the immune response of helper T cells (Th2). Methods 10 C 57 mice infected with cercariae of Schistosoma japonicum were set up as normal control group. After 8 weeks of infection, the mice were killed and the spleen was taken to prepare lymphocyte suspensions, the CD8a antibody was labeled with phycoerythrin (PE) and the IL-4 antibody was stained with phycocyanin (APC). The expression of cytokines in 10 normal C57 mice was detected by flow cytometry. The spleen of 10 normal C57 mice was taken to make lymphocyte suspension and stimulated by Sj FBPA protein in vitro. Results the expression of IL-4,CD8a in spleen cells was (22.39 鹵5.93)% and (11.34 鹵2.15)% respectively after 8 weeks of schistosomiasis infection in mice, and the expression of IL-4,CD8a in splenocytes of normal control group was (1.39 鹵0.52)%. The expression of IL-4,CD8a was (14.14 鹵1.16)% and (8.99 鹵0.81)%, respectively. The expression of IL-4,CD8a was (1.86 鹵0.93)% and (1.69 鹵0.56)% in the normal control group, respectively.
